NSAIDs and Cancer Resolution: New Paradigms beyond Cyclooxygenase

Oluwafunke R. Kolawole et al.

Summary: Acute inflammation is an adaptive host defense mechanism, but unresolved or chronic inflammation can lead to various maladies, including cancer. Nonsteroidal anti-inflammatory drugs (NSAIDs) such as aspirin have been shown to reduce the risk and mortality of many cancers. Besides targeting cyclooxygenase-2 (COX-2), NSAIDs also affect other important targets, and specialized pro-resolving mediators (SPMs) promote the resolution of inflammation and tissue healing, showing potential in inhibiting cancer growth and metastasis.

Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Repurposing of Medicines in the EU: Launch of a Pilot Framework

Charlotte Asker-Hagelberg et al.

Summary: Drug repurposing is the process of finding new uses for existing medicines outside of their original approved indications. The COVID-19 health crisis has highlighted the importance of repurposing drugs to provide early access to treatment for patients. In 2019, a virtual observatory group was established by the Pharmaceutical Committee to support non-profit stakeholders in generating data for new therapeutic uses of authorized medicines.

Repurposing - second life for drugs

Porkodi Ayyar et al.

Summary: Drug repurposing aims to find new indications for existing drugs as a solution to the challenges in new drug development. The paradigm shift from traditional drug discovery to drug repurposing is driven by high costs, long timelines, low success rates, regulatory hurdles, and revenue loss from patent expiration and generic competition. Drug repurposing strategies offer hope in addressing these challenges.

Pirfenidone Sensitizes NCI-H460 Non-Small Cell Lung Cancer Cells to Paclitaxel and to a Combination of Paclitaxel with Carboplatin

Helena Branco et al.

Summary: This study investigated the potential of using Pirfenidone to enhance the sensitivity of non-small cell lung cancer (NSCLC) cells to chemotherapy. The results showed that Pirfenidone increased the effectiveness of paclitaxel treatment on NSCLC cells without increasing toxicity in non-tumorigenic cells. This research suggests the possibility of repurposing Pirfenidone in combination with chemotherapy for treating NSCLC.

Aspirin Suppressed PD-L1 Expression through Suppressing KAT5 and Subsequently Inhibited PD-1 and PD-L1 Signaling to Attenuate OC Development

Xiyun Xiao et al.

Summary: This study investigated the role of aspirin (ASP) in ovarian cancer (OC) treatment. The results showed that ASP inhibited the proliferation of OC cells and suppressed tumor growth in mice. ASP regulated PD-L1 expression through inhibiting KAT5-mediated epigenetic regulation, which in turn suppressed the PD-1/PD-L1 signaling and attenuated tumor growth. ASP may be a promising adjuvant in OC immunotherapy.

Hyaluronic acid (HA)-coated naproxen-nanoparticles selectively target breast cancer stem cells through COX-independent pathways

Eva Espinosa-Cano et al.

Summary: This study investigated the use of hyaluronic acid-coated naproxen polymeric nanoparticles targeting cancer stem cells in metastatic breast cancer. The HA-coating improved drug release control, enhanced hemocompatibility, and displayed strong CSC-targeting efficacy in breast cancer cells. The nano-formulated NAP significantly reduced cell viability and migration, possibly through inducing apoptosis via a GSK-3 beta-related pathway, suggesting potential for improving the treatment of advanced breast cancer.
