Mendelian randomization analyses for PCOS : evidence, opportunities, and challenges

Congying Wang et al.

Summary: Mendelian randomization (MR) assesses causal associations between PCOS risk and related exposure factors through genetic variation. This technology has provided evidence of causal associations of various factors such as anti-Mullerian hormone (AMH) levels, sex hormone-binding globulin (SHBG) levels, menopause age, adiposity, insulin resistance (IR), depression, breast cancer, etc., with PCOS.

Kappa-Opioid Receptor Blockade Ameliorates Obesity Caused by Estrogen Withdrawal via Promotion of Energy Expenditure through mTOR Pathway

Amparo Romero-Pico et al.

Summary: Weight gain is a common symptom of decreased estradiol levels due to menopause or surgical ovariectomy. This study shows that the kappa-opioid receptor system plays a role in mediating weight gain associated with estradiol withdrawal. It also suggests that the hypothalamic mTOR pathway is involved in regulating weight and adiposity in ovariectomized mice.

Liraglutide 3 mg on weight, body composition, and hormonal and metabolic parameters in women with obesity and polycystic ovary syndrome: a randomized placebo-controlled-phase 3 study

Karen E. Elkind-Hirsch et al.

Summary: This study aimed to evaluate the efficacy and safety of the GLP-1 analog liraglutide 3 mg (LIRA 3 mg) in reducing body weight and hyperandrogenism in women with obesity and polycystic ovary syndrome (PCOS). The results showed that LIRA 3 mg was superior to placebo in reducing body weight, lowering androgen levels, and improving cardiometabolic parameters.

11-Oxyandrogens in Adolescents With Polycystic Ovary Syndrome

Anya E. Taylor et al.

Summary: This study investigated the role of 11-oxyandrogens in the diagnosis and clinical comorbidities of PCOS, as well as their response to COCP or metformin therapy. The results showed that untreated PCOS patients had higher levels of 11-OHA4 and 11-OHT, and testosterone was a better predictor of PCOS than 11-oxyandrogens. Among untreated PCOS patients, all three 11-oxyandrogens were correlated with hirsutism severity. COCP treatment was associated with lower levels of 11-KT, while metformin treatment had no effect on 11-oxyandrogens. Further research is needed to determine the causal relationship, metabolic outcomes, and clinical utility of measuring these androgens in PCOS.

Humanin Alleviates Insulin Resistance in Polycystic Ovary Syndrome: A Human and Rat Model-Based Study

Yingying Wang et al.

Summary: Humanin concentrations in the follicular fluid were significantly lower in PCOS patients with IR than those without IR. Supplementing with humanin analogue (HNG) attenuated increases in fasting plasma glucose and fasting insulin levels in rats with PCOS, and improved phosphorylation of key proteins in granulosa cells. The study suggests potential therapeutic use of HNG in treating PCOS.
