RNA-responsive elements for eukaryotic translational control

Summary: The use of eukaryotic toehold switches (eToeholds) allows for efficient induction of gene translation in eukaryotic cells, capable of distinguishing the presence of RNA transcripts under various circumstances. Through optimization of RNA annealing, up to 16-fold induction of transgene expression was achieved.

Programmable deletion, replacement, integration and inversion of large DNA sequences with twin prime editing

Summary: The twin prime editing (twinPE) method described in this study allows for programmable replacement or excision of DNA sequences at endogenous human genomic sites without the need for double-strand DNA breaks (DSBs). When combined with a site-specific serine recombinase, twinPE enables targeted integration of gene-sized DNA plasmids (>5,000 bp) and targeted sequence inversions of 40 kb in human cells. TwinPE expands the capabilities of precision gene editing and may synergize with other tools for the correction or complementation of large or complex human pathogenic alleles.

Engineering synthetic RNA devices for cell control

Summary: The versatility of RNA as a substrate for engineering biological systems lies in its ability to sense and interact with various molecules, proteins, and nucleic acids while encoding genetic instructions. Synthetic RNA devices, through integrating sensing, processing, and actuation of signals, have been developed for controlling cell behavior. Recent advances have overcome challenges in expanding analyte sensing range and adding new mechanisms of action, leading to the maturation of synthetic RNA-based devices. These devices find applications in biomedical research and biomanufacturing, and hold promise for novel therapeutics.

Cryo-EM structures of full-length Tetrahymena ribozyme at 3.1 Å resolution

Summary: This study presents cryo-EM structures of the Tetrahymena ribozyme in substrate-free and bound states, revealing new peripheral helical structures and kissing loop pseudoknots. The global architecture is nearly identical, with significant conformational changes and localized shifts in the internal guide sequence and guanosine binding site upon RNA substrate binding. These results provide a long-sought structural view of a paradigmatic RNA enzyme and signal a new era for cryo-EM-based study of structure-function relationships in ribozymes.

Synthesis of RNA-based gene regulatory devices for redirecting cellular signaling events mediated by p53

Summary: This study developed an aptazyme switch that could sense wild-type p53 protein and flexibly regulate downstream gene function, achieving targeted killing effect on tumor cells in combination with other synthetic biological tools. The aptazyme successfully inhibited tumor growth by specifically recognizing p53 and provided a modular strategy for binding to cellular proteins.
