EGFR Ligand Clustering on E2 Bionanoparticles for Targeted Delivery of Chemotherapeutics to Breast Cancer Cells

Summary: In this study, SpyTag peptide was fused to the surface of engineered E2 nanocages to enable tunable nanocage decoration and effective E2 cell targeting using a variety of SpyCatcher fusion proteins. The core of the E2 nanocage incorporated four phenylalanine mutations for hydrophobic drug loading and pH-responsive release. By functionalizing the nanocage surface with a highly cell-specific EGFR-targeting protein conjugate, efficient drug loading and targeted cell death in inflammatory breast cancer cells were achieved. This engineered protein nanocage construct demonstrates versatility for cargo delivery.

However, the commercial success at industrial scale is still inadequate due to irregular biomass supply chain, market uncertainties, and scale-up challenges. Global research efforts by public and private sectors are underway to achieve deeper market penetration for the commercial success of lignocellulosic biorefineries.

This review discusses the latest methods and technologies in biomass pretreatment, as well as genetic engineering techniques for developing advanced strategies in fermentation processes. The aim is to bridge the gap in developing economically feasible lignocellulosic products and chemicals using biorefining technologies.

Pathways of lignocellulosic biomass deconstruction for biofuel and value-added products production

Summary: This paper addresses the gaps in the existing literature review works by comprehensively evaluating different pretreatment methods and applications of lignocellulosic biomass (LCB) and incorporating techno-economic considerations. The review finds that physical and chemical pretreatment methods, although effective, are highly dependent on energy and chemicals, producing waste and inhibitory compounds. Biological pretreatment can address these challenges but has longer incubation times. Integrated/coupling methods have gained popularity for improving chemical yield but come at a high cost of operation. Process integration and optimization are needed to make pretreatment processes more techno-economically feasible.

Bacillus subtilis as a robust host for biochemical production utilizing biomass

Summary: Bacillus subtilis is considered a suitable host for biochemical production due to its excellent growth and bioresource utilization characteristics, as well as the adaptability of endogenous metabolic pathways and heterologous pathways. Its efficient protein secretion system makes it suitable for producing various biochemicals, while metabolic and genetic engineering techniques have improved production titers and product selectivity. This review highlights recent progress in utilizing robust B. subtilis-based host systems for biomass conversion and biochemical production, suggesting promising prospects for such host systems.

Cell-surface binding domains from Clostridium cellulovorans can be used for surface display of cellulosomal scaffoldins in Lactococcus lactis

Summary: Engineering synthetic scaffoldins derived from C. cellulovorans cellulosome components successfully introduced them into L. lactis, with a focus on their potential cell-surface anchoring capabilities. Among the engineered scaffoldins, only those carrying three EngE SLH modules were able to bind to the surface of L. lactis, despite the absence of the conserved TRAE motif typically involved in binding with secondary cell wall polysaccharides. These synthetic scaffoldins could potentially be used for assembling secreted or surface-displayed designer cellulosomes in L. lactis.

Engineering a Blue Light Inducible SpyTag System (BLISS)

Summary: The SpyCatcher/SpyTag protein conjugation system has gained popularity due to its fast kinetics and high yield, and by integrating light dependency into this system, a new opportunity for biofabrication and optogenetics applications has been created. This system allows for spatial and temporal control of the conjugation event and can be instantaneously quenched by removing light.
