Insights into the Polymorphic Structures and Enantiotropic Layer-Slip Transition in Paracetamol Form III from Enhanced Molecular Dynamics

Richard S. Hong et al.

Summary: Reversible temperature-mediated solid phase changes, known as enantiotropic transformations, occur in many molecular crystals due to entropic contributions. Understanding and predicting these transformations are crucial, and molecular simulations were used in this study to elucidate the mechanism behind the enantiotropic layer-slip phase transformation in paracetamol form III. The results suggest that the transformation is driven by localized and dynamic disorder within the structure, rather than a perfect crystal-to-crystal transition. The orthorhombic form III-o structure exists as an entropy-stabilized collective ensemble average of various misaligned layer-slipped structures.

Summary: The behavior of many materials is strongly influenced by the mechanical properties of hard phases, which can be deliberately introduced for reinforcement or may be present as deleterious precipitates. Studying these phases is hindered by their small sizes and lack of bulk ductility at room temperature. Researchers have turned to small-scale testing to study deformation mechanisms of complex crystal structures, using nanoindentation, microcompression, electron microscopy, and orientation measurements for interpretation.
