Evaluation of the kinetics of unseeded batch cooling crystallization using population balance modeling: Sucrose and KNO3 case studies

Seunghwan Kim et al.

Summary: A facile method for estimating the kinetics of unseeded batch crystallization was developed through mass and average size measurements. The population balance equation (PBE) was reformulated as a function of both mass and average size to determine mass balance and obtain crystallization kinetics. Inorganic and organic compounds were evaluated, and the method allowed for the extraction of kinetic parameters for crystal nucleation and growth. The predicted crystallization behaviors were validated with experiments, showcasing the usefulness of the developed method for evaluating lumped kinetic parameters of unseeded batch crystallizations.

The unexpected dominance of secondary over primary nucleation

Johannes Hoffmann et al.

Summary: Controlling particle formation in industrial crystallization processes is still a challenge. Secondary nucleation is identified as the dominant nucleation mechanism, and enantioselective secondary nucleation can be used to control the chirality of the product, making antisolvent crystallization a viable crystallization-enhanced deracemization technique.

Stochastic shelf-scale modeling framework for the freezing stage in freeze-drying processes

Leif-Thore Deck et al.

Summary: Freezing and freeze-drying processes are commonly used in pharmaceutical formulations to improve stability. However, batch heterogeneity can cause process failure. In this study, a modeling framework for large-scale freezing processes was developed and an open-source implementation was published. The model couples heat transfer with ice nucleation kinetics and showed how ice nucleation leads to heterogeneity. Various cooling protocols were investigated, and holding schemes were found to have similar solidification times as controlled nucleation, suggesting a potential pathway for process optimization.

Secondary Nucleation by Interparticle Energies. III. Nucleation Rate Model

Byeongho Ahn et al.

Summary: A nucleation rate model for secondary nucleation caused by interparticle energies (SNIPEs) is derived and validated through numerical verification and experimental validation. The model shows that SNIPE mechanism enhances primary nucleation by reducing the energy barrier and critical nucleus size due to interparticle interactions. The model is also consistent with other secondary nucleation rate models and has better parameter estimation.

Modeling of nucleation time distribution using Laplace transform of activity density function - Stochastic heterogeneous nucleation

Noriaki Kubota

Summary: This paper investigates heterogeneous nucleation in small samples induced by active sites under isothermal conditions. The study reveals that increasing the number or activity of active sites leads to easier or earlier occurrence of heterogeneous nucleation. The nucleation time varies significantly among different samples and shows a wide distribution. By using mathematical models and data analysis, the paper derives equations for the nucleation time distribution and calculates the activity density and number distribution of active sites. The results indicate that the nucleation time distribution is mainly governed by a small number of highly active sites, while other less active sites have no impact on nucleation time.

Characterization of a small-scale crystallizer using CFD simulations and X-ray CT measurements

Ramona Achermann et al.

Summary: Although small-scale crystallizers have gained increasing importance, their hydrodynamic properties have not been thoroughly studied. This work characterizes an intensively stirred small-scale crystallizer and analyzes its fluid mechanics behavior. Single and multiphase computational fluid dynamics simulations show significant differences in the mixing characteristics of two types of stirrers, leading to variations in solids distribution. X-ray computed tomography measurements confirm spatial variations in solid distribution and highlight the potential errors in assuming a homogeneous suspension when modeling crystallization processes.

Online Monitoring of the Concentrations of Amorphous and Crystalline Mesoscopic Species Present in Solution

Byeongho Ahn et al.

Summary: This study successfully used depolarized light sheet microscopy to achieve time-resolved measurements of concentrations of amorphous and crystalline mesospecies in solutions, demonstrating the importance of mesospecies formation and the reliability of the measurement method under different experimental conditions. The results revealed that the direct measurement of crystal nucleation rates in solutions at different levels of supersaturation showed reasonable agreement with the predictions of classical nucleation theory.

Stochastic ice nucleation governs the freezing process of biopharmaceuticals in vials

Leif-Thore Deck et al.

Summary: This study investigates the freezing process of pallets used in biopharmaceuticals and compares it with engineering run data from the Janssen COVID-19 vaccine. The researchers found that stochastic ice nucleation plays a crucial role in determining process duration and batch heterogeneity. Understanding the ice nucleation kinetics of the formulation is important for identifying suitable freezing process conditions.

On the linear and integral models for metastable zone width determined by solution cooling method

Keshra Sangwal

Summary: Based on classical nucleation theory, final equations regarding the metastable zone width were derived using three deterministic approaches, which were then applied to analyze experimental data from different systems. The analysis compared and discussed trends in nucleation kinetic parameters values obtained from the deterministic integral nucleation model and the conventional stochastic theory. Results showed that the solute-supply approach could explain the dependency of solutions' Delta T-m values on cooling rate at different saturation temperatures and estimate nucleation parameters, while relatively low values of the solid-solution interfacial solution gamma(eff) suggested crystallization by heterogeneous nucleation. The nature of nucleation encountered in the MSZW measurements by the polythermal method was also discussed.

A Review of Experimental Methods for Nucleation Rate Determination in Large-Volume Batch and Microfluidic Crystallization

Cedric Devos et al.

Summary: Determination of the experimental nucleation rate for crystallization in solution is crucial for improving industrial crystallization processes and understanding nucleation mechanisms. While large-volume batch crystallizer models have been widely studied in the past, recent developments in microfluidic platforms have opened new pathways for understanding nucleation on a molecular level.

Influence of Volume on the Nucleation of Model Organic Molecular Crystals through an Induction Time Approach

Isaac Jerome C. Dela Cruz et al.

Summary: The analysis revealed that a reduction in sample volume leads to an exponential increase in induction times. Using volume reduction as an approach to nanocrystal development and polymorph access, it was found that acetaminophen systems did not show polymorphism variation across all volumes, while glycine systems shifted towards gamma-glycine nucleation at smaller volume sizes.

Numerical Simulations of Seeded Batch Crystallization Demonstrating the Effect of Stochastic Nucleation on Crystal Product Quality

Joi Unno et al.

Summary: Numerical simulations were used to study the impact of stochastic nucleation on crystal quality parameters, revealing differences in product quality under certain conditions. Stochastic primary nucleation was identified as the main source of discrepancies between deterministic and stochastic simulations, while scaling up the crystallizer was found to reduce fluctuations caused by stochastic nucleation.
