A Heme-Acquisition Protein Reconstructed with a Cobalt 5-Oxaporphyrinium Cation and Its Growth-Inhibition Activity Toward Multidrug-Resistant Pseudomonas aeruginosa

Summary: Using artificial hemes to reconstruct natural heme proteins enhances their bioactivity. Researchers synthesized metal 5-oxaporphyrinium cations and successfully incorporated cobalt 5-oxaporphyrinium cation into heme-acquisition protein secreted by Pseudomonas aeruginosa. The hypothesis is that the oxaporphyrinium cation binds strongly to the HasA-specific outer membrane receptor, preventing heme acquisition. In fact, the reconstructed HasA inhibits the growth of Pseudomonas aeruginosa and multidrug-resistant P. aeruginosa.
