Methylene blue and ascorbate interfere with the accurate determination of the kinetic properties of IDO2

Summary: This study compared IDO1 and IDO2 from different species and found that IDO2 is more stable than IDO1 in the absence of a reductant, engaging in multiple turnovers of the reaction. The enzymatic activity of IDO2 is significantly affected by the use of different reductants, suggesting that IDO2 may also participate in the metabolism of Trp in vivo.

Direct RBS Engineering of the biosynthetic gene cluster for efficient productivity of violaceins in E. coli

Summary: A direct RBS engineering strategy was proposed to improve the fermentation titers of the violacein biosynthetic gene cluster, achieving a 2.41-fold increase in production compared to the starting strain. By optimizing batch-fermentation parameters, a maximum yield of violaceins was reached at higher temperatures, providing potential for greater improvement in product yields.

Diversity of the Tryptophanase Gene and Its Evolutionary Implications in Living Organisms

Summary: Tryptophanase encoded by the gene tnaA is a pyridoxal phosphate-dependent enzyme that catalyses the conversion of tryptophan to indole, commonly used as a signalling molecule by microbes. While tnaA is commonly found in various Gram-negative bacteria, it is rare in Gram-positive bacteria and archaea. Its presence in eukaryotes, particularly protozoans and marine organisms, highlights its importance in the animal kingdom. This study documents the distribution and acquisition of tnaA among different taxonomic groups, providing insights into its role among various organisms.

Tryptophanase gene deficiency improves the application of dioxygenase to 3-(2-hydroxyethyl)catechol production

Summary: In this study, a biocatalytic method for producing HEC from 2-phenylethanol was established using an isolated strain. It was found that disruption of the tryptophanase gene in E. coli could enhance HEC production by repressing the generation of blue dyes, which likely inhibited HEC production.

Parallel evolution of trehalose production machinery in anhydrobiotic animals via recurrent gene loss and horizontal transfer

Summary: Trehalose, a versatile non-reducing sugar, is used as a potent protectant against environmental stresses and blood sugar in certain animal groups. Despite the unidentified trehalose biosynthesis genes in the large majority of metazoan phyla, bifunctional TPS-TPP genes have been identified across various taxa. Phylogenetic analyses revealed the acquisition of 'pan-metazoan' genes in early metazoan ancestors, indicating parallel evolution of trehalose synthesis through recurrent loss and horizontal gene transfer.
