Recent Advances in Plasmonic Nanostructures Applied for Label-free Single-cell Analysis

Single-cell analysis is necessary to characterize cell-to-cell differences during biochemical processes. Plasmonic nanostructures, utilizing unique localized surface plasmon resonance and Mie scattering, provide stable scattering signals for dynamic trace and biological imaging of single living cells, avoiding photobleaching and autofluorescence. Various techniques like surface-enhanced Raman scattering, surface-enhanced Infrared absorption spectroscopy, and dark-field microscopy are used for label-free detection and monitoring of target cells at the single-cell level, demonstrating the potential of plasmonic probes for single-cell analysis.

Simultaneous Visualization of Dual Intercellular Signal Transductions via SERS Imaging of Membrane Proteins Dimerization on Single Cells

Summary: A highly sensitive and specific surface-enhanced Raman scattering (SERS) strategy was proposed to simultaneously image dual homodimerizations of membrane proteins on single live cells. Networking of AuNPs-based dual-recognition probes (dual-RPs) and SERS tags via proximity ligation-assisted catalytic hairpin assembly (CHA) was used to enhance the SERS effect. This strategy provides a good method for high-sensitivity monitoring of membrane proteins dimerizations-based multiple intercellular signal transductions in a natural and complex cellular microenvironment.

Using the DNA origami structure, single-molecule SERS measurements of three dyes, cytochrome c, and horseradish peroxidase proteins have been successfully demonstrated.
