Structure-function studies can improve binding affinity of cohesin-dockerin interactions for multi-protein assemblies

Summary: The cellulosome is a complex multi-enzyme structure secreted by anaerobic microorganisms for efficient degradation of lignocellulosic substrates. It consists of catalytic and non-catalytic components that are assembled through protein-protein interactions. The structure of an adaptor scaffoldin-borne ScaH Doc in complex with the Coh from anchoring scaffoldin ScaE was described, revealing a dual-binding mode interaction and identifying key residues for Coh recognition. Rational protein engineering was used to improve the affinity between the two modules, showing the potential for tailored Coh-Doc interactions in bioengineering applications.

Microscale Colocalization of Cascade Enzymes Yields Enhancement

Summary: This study establishes a cascade system using reversible DNA-directed colocalization of enzymes on microspheres to directly evaluate the colocalization effect. In a regime of highly dilute microspheres of particular sizes, the colocalized cascade shows enhanced activity compared to the freely diffusing cascade. The findings provide theoretical support for the rational design of highly efficient multienzyme catalysts.

Design of functional hydrogels using smart polymer based on elastin-like polypeptides

Summary: In this study, we designed and synthesized genetically encoded materials called elastin-like polypeptides (ELPs) that can undergo temperature-induced phase transition and photo-crosslinking due to modification with methacrylate groups. The degree of methacrylate functionalization can be controlled by adjusting the reactant ratio. By combining hydrogel fabrication strategy, the methacrylated ELPs (ELP-MA) can be engineered into different physical forms such as coacervates and hydrogels, functioning as light transmittance regulators and elastic adhesives. The physical properties of the hydrogels can be controlled by the degree of methacrylation and ELP-MA concentration. The resulting ELP-MA hydrogels showed good biocompatibility when implanted subcutaneously in mice.
