Mitochondria ROS and mitophagy in acute kidney injury

Lianjiu Su et al.

Summary: Mitophagy is a crucial mitochondrial quality control mechanism that removes damaged mitochondria and reactive oxygen species (ROS) production. The relationship between mitochondrial oxidative stress, ROS production, and mitophagy is closely intertwined and involved in various pathological conditions of acute kidney injury (AKI). The complex process of mitophagy in mammals involves multiple pathways and its interaction with different types of cell death, such as apoptosis, pyroptosis, and ferroptosis in kidney injury, remains unclear. This review discusses recent advances in understanding the relationship between ROS and mitophagy, the different pathways of mitophagy, the interplay between mitophagy and cell death, and their significance in the pathogenesis of AKI.

Mitochondrial Ferritin Deficiency Promotes Osteoblastic Ferroptosis Via Mitophagy in Type 2 Diabetic Osteoporosis

XinDong Wang et al.

Summary: The incidence of type 2 diabetic osteoporosis (T2DOP) is rapidly increasing and its specific mechanism remains unclear. Studies have shown a relationship between iron overload and T2DOP. Mitochondrial ferritin (FtMt) may inhibit ferroptosis in osteoblasts by reducing oxidative stress caused by excess ferrous ions, while FtMt deficiency induces mitophagy in the pathogenesis of T2DOP. FtMt could potentially serve as a target for T2DOP therapy.

Targeting Mitochondrial COX-2 Enhances Chemosensitivity via Drp1-Dependent Remodeling of Mitochondrial Dynamics in Hepatocellular Carcinoma

Lin Che et al.

Summary: Translation: New therapeutic strategies are needed to improve the anti-cancer effect for hepatocellular carcinoma (HCC). This study explores the involvement of mitochondrial translocation of cyclooxygenase-2 (COX-2) in mitochondrial dynamics and sensitization of HCC cells to anti-tumor therapy. The findings suggest that targeting mitochondrial COX-2 could be a potential therapeutic approach for HCC and other cancers with high expression of COX-2.

Selenium inhibits ferroptosis and ameliorates autistic-like behaviors of BTBR mice by regulating the Nrf2/GPx4 pathway

Hongmei Wu et al.

Summary: The study found that sodium selenite (Se) has a positive effect on autism spectrum disorder (ASD) behaviors and inhibits ferroptosis. It acts by modulating the Nrf2/GPx4 signaling pathway.

β-amyloid protein induces mitophagy-dependent ferroptosis through the CD36/PINK/PARKIN pathway leading to blood-brain barrier destruction in Alzheimer's disease

Jianhua Li et al.

Summary: The dysfunction of pericytes in the blood-brain barrier (BBB) is associated with the onset of Alzheimer's disease (AD), and CD36 plays a role in the disruption of the BBB. Beta-amyloid protein has a negative impact on pericytes and may cause BBB damage by inducing mitophagy-dependent ferroptosis.

Cystine transporter SLC7A11/xCT in cancer: ferroptosis, nutrient dependency, and cancer therapy

Pranavi Koppula et al.

Summary: The overexpression of SLC7A11 promotes tumor growth while causing significant metabolic reprogramming costs for cancer cells. Cancer cells with high expression of SLC7A11 exhibit glucose- and glutamine-dependency, presenting potential metabolic vulnerabilities for therapeutic targeting.

Role of ferroptosis in neurological diseases

Min-Yi Yao et al.

Summary: Ferroptosis, a newly identified form of nonapoptotic regulated cell death characterized by iron-dependent accumulation of lipid peroxides causing oxidative stress and cell death, has been found to play an essential role in the pathology of neurological diseases. Recent studies highlight the potential of ferroptosis-based mechanisms as new therapeutic targets for neurological disorders.

A preliminary study on the mechanism of the neurosteroid-mediated ionotropic receptor dysfunction in neurodevelopmental toxicity induced by decabromodiphenyl ether

Bo Qian et al.

Summary: The neurodevelopmental toxicity mechanism of decabromodiphenyl ether (BDE209) is not fully understood, but recent evidence indicates that neurosteroids disorders may play a key role. Exposure to BDE209 during pregnancy and lactation was found to impair offspring's learning and memory abilities and disrupt neuronal structure. This effect may be mediated through dysfunction of ionotropic receptors by neurosteroids.

Organophosphate flame retardants and diesters in the urine of e-waste dismantling workers: associations with indoor dust and implications for urinary biomonitoring

Rui-Xin Qin et al.

Summary: Indoor dust ingestion is a major pathway for human exposure to organophosphate flame retardants (PFRs), and there are significant correlations between PFR levels in dust and urinary concentrations of diesters (DAPs). This suggests potential sources and pathways of exposure to these chemicals.
