Summary: Thirteen designed and synthesized small molecule compounds showed significant cytotoxicity against tumor cell lines, with compound 5l displaying the highest potency and significant caspase activation activity. It was found that compound 5l may act as a potent allosteric inhibitor of procaspase-3 through chelation of the inhibitory zinc ion.

Investigation of quinolone-tethered aminoguanidine as novel antibacterial agents

Summary: This study optimizes the previous hit to discover quinolone-tethered aminoguanidine compounds with good antitubercular activity, including compounds 7f and 8e with submicromolar MIC90 values. These compounds show a fivefold increase in antitubercular activity compared to the previous study. The compounds exhibit poor antibacterial activities against ESKAPE pathogens but no cytotoxicity effects against noncancerous human cell lines. In silico studies predict good solubility, no inhibitory effect on cytochrome P450 isoenzymes, and non-interference in multiple assays for compound 7f.

Tapping into the antitubercular potential of 2,5-dimethylpyrroles: A structure-activity relationship interrogation

Summary: The exploration around the 2,5-dimethylpyrrole scaffold of antitubercular hit compound 1 has led to the discovery of new derivatives that are active against Mycobacterium tuberculosis and multidrug-resistant clinical isolates. Some of these derivatives showed promising cytotoxicity profiles and were effective in inhibiting the growth of intracellular mycobacteria.
