AAZTA-Derived Chelators for the Design of Innovative Radiopharmaceuticals with Theranostic Applications

AAZTA-derived chelators, especially suitable for short half-life radioelements (i.e., Ga-68), show great potential in the design, synthesis, and applications in nuclear medicine/pharmacy field.

The critical role of ligand topology: strikingly different properties of Gd(iii) complexes with regioisomeric AAZTA derivatives

Summary: This study highlights the importance of ligand topology in the properties of Gd(iii) complexes relevant in the field of magnetic resonance imaging (MRI). By comparing two Gd(iii) complexes, it was found that they have different hydration numbers, stability, kinetic inertness, and water exchange rates. Differences were also observed in their interaction and relaxivity with human serum albumin.

In targeting integrin alpha(v)beta(3) receptors on cancer cells for molecular imaging and diagnostic applications, multimeric radioligands have shown superiority over monomers. Structural characteristics, binding avidity, pharmacodynamic properties, and PET imaging features of these multimeric ligands have been comprehensively analyzed and discussed in this review.
