We have developed a class of amphiphilic drug complexes by programming hydrophobic drug patterns on DNA frameworks. We have investigated the impact of these drug patterns on cellular uptake efficiency and drug potency, and have achieved enhanced cytotoxicity against tumor cells through the use of an asymmetric drug pattern.
