Rational Design, Synthesis and Evaluation of Novel Rodanine Derivatives for Antihyperglycemic Activity

Summary: A new series of rhodanines with phenyl substitution were designed and synthesized for their potential antihyperglycemic activity targeting the PPAR-gamma receptor. Rational design and molecular modeling studies were performed, and the synthesized compounds were confirmed by structural analysis. In vitro and in vivo assays were conducted to evaluate the glucose uptake and antihyperglycemic activity of the compounds. Some compounds showed promising activity, with compound B5 exhibiting superior antihyperglycemic activity compared to the standard drug pioglitazone.

5-Aryl-furan derivatives bearing a phenylalanine- or isoleucine-derived rhodanine moiety as potential PTP1B inhibitors

Summary: Two series of 5-aryl-furan derivatives with a phenylalanine- or isoleucine-derived rhodanine moiety were identified as competitive PTP1B inhibitors. Compound 5g showed the best PTP1B inhibitory potency and CDC25B inhibitory potency, with the introduction of a sec-butyl group at the 2-position of the carboxyl group significantly improving the PTP1B inhibitory activity.
