Microenvironmental effects can masquerade as substrate channelling in cascade biocatalysis

Walaa Abdallah et al.

Summary: Natural cascades often use spatial organization to introduce beneficial substrate channeling mechanisms, which has been widely adopted in engineered multienzyme cascades. Utilizing new molecular scaffolds now allows testing the effects of spatial organization on cascade kinetics, but these scaffolds can also change the microenvironment experienced by the assembled enzymes, complicating kinetic analysis.

Microscale Colocalization of Cascade Enzymes Yields Enhancement

Yan Xiong et al.

Summary: This study establishes a cascade system using reversible DNA-directed colocalization of enzymes on microspheres to directly evaluate the colocalization effect. In a regime of highly dilute microspheres of particular sizes, the colocalized cascade shows enhanced activity compared to the freely diffusing cascade. The findings provide theoretical support for the rational design of highly efficient multienzyme catalysts.

How Pore Architecture Regulates the Function of Nanoscale Protein Compartments

Nuren Tasneem et al.

Summary: Self-assembling proteins can form well-defined architectures at the nanoscale, acting as semipermeable barriers for spatial separation and organization of biochemical processes. Engineering the structure of protein compartments allows for tuning their functionality, enabling non-native applications such as artificial nanoreactors. This review provides insights into how protein structure determines the porosity and function of compartments, with a focus on recent structural data obtained by cryo-EM and X-ray crystallography. The authors suggest that these structural insights can guide the design of artificial protein compartments with tunable porosity and function.

Modular polyketide synthase contains two reaction chambers that operate asynchronously

Saket R. Bagde et al.

Summary: Type I modular polyketide synthases are enzymes that synthesize a variety of polyketide natural products by performing polyketide chain extension and b-keto group modification reactions. The structures of Lsd14 polyketide synthase revealed the positioning of constituent domains, rearrangement depending on the reaction cycle step, and specific interactions formed between domains. Lsd14 contains two reaction chambers, but only one chamber has the full complement of catalytic domains producing the polyketide product at any given time.
