Peptide-Decorated DNA Nanostructures Promote Site-Specific Hydroxyapatite Growth

Alexander L. Danesi et al.

Summary: The study demonstrates that DNA nanostructures can promote site-specific calcium phosphate formation under mineralizing conditions, resulting in amorphous calcium phosphate or hydroxyapatite. This finding sheds light on the mechanisms of crystal nucleation and growth in mineralized tissues, providing insights for mineralized tissue regeneration strategies.

Modular Synthesis of Trifunctional Peptide-oligonucleotide Conjugates via Native Chemical Ligation

Mohadeseh Dastpeyman et al.

Summary: Cell penetrating peptides (CPPs) are efficient vectors for intracellular delivery of biologically active agents like therapeutic antisense oligonucleotides (ASOs). Conjugating ASOs to CPPs through native chemical ligation (NCL) significantly improves their cellular permeability and therapeutic efficacy. This study demonstrates that chemo-selective NCL is a superior strategy for synthesizing bi-/trifunctional CPP-ASO conjugates with high efficiency.

A Versatile Solid-Phase Approach to the Synthesis of Oligonucleotide Conjugates with Biodegradable Hydrazone Linker

Mariya I. Meschaninova et al.

Summary: A convenient solid-phase approach for synthesizing 5'-conjugates of oligonucleotides with biodegradable pH-sensitive hydrazone covalent bonds is proposed in this study. The proof-of-principle of this approach was demonstrated by synthesizing lipophilic conjugates of modified siRNA and non-coding RNAs imported into mitochondria. This method has the potential to be extended for synthesizing pH-sensitive conjugates of oligonucleotides with ligands of different nature.

Janus Nanosheets with Face-Selective Molecular Recognition Properties from DNA-Peptide Conjugates

Shine K. Albert et al.

Summary: The study demonstrates the fabrication of Janus nanosheets from molecular building blocks composed of DNA and peptides, and reveals the formation of nanosheets with distinct DNA and peptide faces. The surprisingly level of structural control is attributed to the exclusive parallel beta-sheet formation of phenylalanine-rich peptides. Furthermore, the predictable and designable peptide interaction is shown to be a promising tool for creating Janus nanostructures with regio-selective and sequence-specific molecular recognition properties.
