Histone methyltransferase SETDB1 inhibits TGF-β-induced epithelial-mesenchymal transition in pulmonary fibrosis by regulating SNAI1 expression and the ferroptosis signaling pathway

Tiantian Liu et al.

Summary: SETDB1 regulates Snai1 expression epigenetically, driving EMT gene reprogramming and ferroptosis in response to TGF-beta. However, the links between epigenetic reprogramming and transcriptional processes in TGF-beta-dependent EMT regulation remain unexplored.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

The emerging roles of circRNAs in cancer and oncology

Lasse S. Kristensen et al.

Summary: Circular RNAs (circRNAs) are a novel class of primarily non-coding RNAs with increasingly recognized roles in cancer development and progression through diverse mechanisms of action. In this review, the authors summarize the current understanding of circRNA biogenesis, regulation, physiological functions and pathophysiological roles in cancer. They also discuss the clinical potential of circRNAs as biomarkers, therapeutic agents and drug targets in oncology, as well as the research controversies, technical issues, and biological knowledge gaps that need to be addressed.

Enhancer of zeste homolog 2 promotes hepatocellular cancer progression and chemoresistance by enhancing protein kinase B activation through microRNA-381-mediated SET domain bifurcated 1

Jingyang Zhou et al.

Summary: This study investigated the role and mechanism of miR-381 in the development and chemoresistance of HCC. It was found that EZH2 suppressed miR-381 expression, leading to increased SETDB1 expression and activation of the AKT pathway, thereby promoting hepatocarcinogenesis and chemoresistance.

PABPC1-induced stabilization of IFI27 mRNA promotes angiogenesis and malignant progression in esophageal squamous cell carcinoma through exosomal miRNA-21-5p

Ying Zhang et al.

Summary: Emerging evidence has shown that dysregulation of RNA-binding proteins is involved in the progression of esophageal squamous cell carcinoma (ESCC). This study aimed to investigate the role of poly (A) binding protein cytoplasmic 1 (PABPC1) in ESCC progression. PABPC1 expression was found to be upregulated in ESCC tissues compared to normal esophageal epithelial tissues. Elevated PABPC1 expression was associated with tumor cell differentiation and poor prognosis. Mechanistically, PABPC1 interacted with eIF4G to regulate the stability of IFI27 mRNA and promote angiogenesis through exosomal miR-21-5p/CXCL10 signaling. These findings suggest that PABPC1 could be a promising therapeutic target for ESCC.

Summary: RNA-binding proteins (RBPs) regulate gene expression by post-transcriptionally influencing various RNA biology processes, and dysregulation of RBPs may lead to human diseases such as cancer. RBPs play critical roles in important cellular phenotypes and treatment strategies for cancer metastasis.

SETDB1 is required for intestinal epithelial differentiation and the prevention of intestinal inflammation

Lea Juznic et al.

Summary: SETDB1 plays a crucial role in intestinal epithelial homeostasis, and its deficiency may lead to defects in differentiation and inflammation in the intestinal epithelium. Rare variants associated with loss of function in SETDB1 were found in some IBD patients, suggesting a potential contribution to the pathogenesis of intestinal inflammation.

circDCUN1D4 suppresses tumor metastasis and glycolysis in lung adenocarcinoma by stabilizing TXNIP expression

Yingkuan Liang et al.

Summary: The aberrant expression of circRNAs plays a role in cancer progression through interactions with RNA-binding proteins (RBPs). A novel circRNA, circDCUN1D4, has been identified to be involved in the metastasis and glycolysis of lung adenocarcinoma (LUAD) by facilitating interactions between HuR and TXNIP proteins. It acts as a scaffold to enhance stability and transportation of TXNIP mRNA, ultimately suppressing metastasis and glycolysis of lung cancer cells in a TXNIP-dependent manner. The downregulated expression of circDCUN1D4 in lymph node metastatic tissues serves as an independent risk factor for the overall survival of LUAD patients.

Epigenetic silencing by SETDB1 suppresses tumour intrinsic immunogenicity

Gabriel K. Griffin et al.

Summary: A CRISPR-Cas9 screen in mouse tumour models treated with immune checkpoint blockade identified SETDB1 as an epigenetic checkpoint protein that suppresses tumour-intrinsic immunogenicity. Loss of SETDB1 leads to derepression of immune-stimulating genes and triggers TE-specific cytotoxic T cell responses, suggesting it as a potential target for immunotherapy in cancer treatment.

The molecular basis of coupling between poly(A)-tail length and translational efficiency

Kehui Xiang et al.

Summary: The study found that mRNA poly(A)-tail length strongly influences translational efficiency (TE) in animal oocytes and early embryos, but this coupling disappears later in development. Overexpressing cytoplasmic poly(A)-binding protein weakened the coupling between tail length and TE, while postembryonic mammalian cell lines had properties preventing strong coupling.

Linking EMT programmes to normal and neoplastic epithelial stem cells

Arthur W. Lambert et al.

Summary: Epithelial stem cells play critical roles in tissue generation, maintenance, and repair, while cancer stem cells are implicated in the growth and recurrence of many carcinomas. The activation of epithelial-mesenchymal transition (EMT) is linked to the formation of both normal and neoplastic stem cells, but the mechanistic basis of this connection remains unclear. Understanding the elusive links between EMT programs and the stem cell state will advance our knowledge of normal stem cell biology and cancer pathogenesis.
