Targeting LAG3/GAL-3 to overcome immunosuppression and enhance anti-tumor immune responses in multiple myeloma

Targeting LAG3 with antibody treatment enhanced T cells proliferation and activities against MM. Combination therapeutic strategies, including blocking LAG3/GAL-3 and multipeptide vaccination, were proposed to improve anti-tumor responses in MM.

PKCα/JZFP64/CSF1 axis resets the tumor microenvironment and fuels anti-PD1 resistance in hepatocellular carcinoma

Summary: This study identified ZFP64 as an upregulated gene in tumor tissues from HCC patients with anti-PD1 resistance. Elevated ZFP64 contributes to anti-PD1 resistance by promoting M2 macrophage polarization and creating an inhibitory tumor microenvironment. The PKC alpha/ZFP64/CSF1 axis plays a critical role in immune evasion and anti-PD1 tolerance. Inhibiting this axis can overcome resistance to anti-PD1 in HCC.

Transcriptome analysis of Homo sapiens and Mus musculus reveals mechanisms of CD8+T cell exhaustion caused by different factors

Summary: This study conducted a transcriptome analysis by integrating exhaustion datasets from multiple diseases in humans and mice to dissect the molecular mechanisms of T cell exhaustion. The study identified a set of overlapping genes in exhausted CD8+ T cells in both species, which were enriched in exhaustion response-related pathways. Furthermore, the study revealed the hub genes involved in exhaustion, as well as the genes significantly correlated with T cell exhaustion across species. Additionally, the study identified two less described genes, CD200R1 and ADGRG1, contributing to T cell exhaustion. Overall, these findings provide insights into the mechanisms of T cell exhaustion and serve as a valuable reference for the immunology community.

Epigenetic modulation of FBW7/Mcl-1 pathway for lung cancer therapy

Summary: Methylation of tumor suppressor genes in human lung cancer can be reversed by DAC, leading to activation of FBW7 and degradation of Mcl-1 protein, thereby inhibiting tumor growth. DAC treatment converts the FBW7 gene from a methylated to an unmethylated form, resulting in increased FBW7 expression and decreased Mcl-1 levels, ultimately suppressing lung cancer growth. Combination therapy with DAC and Venetoclax shows strong synergistic effects against lung cancer without causing normal tissue toxicity.

Summary: CD39, expressed by tumor-infiltrating lymphocytes, is a marker for identifying tumor-reactive T cells with strong antitumor activity. Constructed HBVs-CAR-T cells and personalized tumor-reactive CD8(+) T cells show increased apoptosis in HCC organoids, with CD39(+) cells demonstrating enhanced antitumor effect despite increased exhaustion status.Triple knockdown of PD-1, Tim-3, and Lag-3 further enhances antitumor activity in CD39(+) CAR-T cells, indicating CD39 as a potential biomarker for immune cell enrichment and prognosis evaluation in HCC immunotherapy.
