Distinct effects of interleukin-6 and interferon-γ on differentiating human cortical neurons

Annie Kathuria et al.

Summary: Translational evidence suggests that cytokines involved in maternal immune activation can harm fetal brain and increase the risk of neuropsychiatric disorders. This study differentiated human pluripotent stem cells to cortical neurons and exposed them to interleukin-6 (IL-6) or interferon-gamma (IFN-gamma). The findings show that IL-6 affects gene expression related to extracellular matrix, actin cytoskeleton, and TGF-beta signaling, while IFN-gamma affects genes related to antigen processing, major histocompatibility complex, and endoplasmic reticulum biology. Mitochondrial respiration and dendritic spine density were also influenced by these cytokines, but pre-treatment with folic acid mitigated their effects.

Wnt5a promotes hippocampal postsynaptic development and GluN2B-induced expression via the eIF2α HRI kinase

Eva Ramos-Fernandez et al.

Summary: The Wnt5a signaling pathway plays a crucial role in neurodevelopment and neuronal maturation by regulating synaptic structures and functions. Specifically, Wnt5a induces changes in PSD length, synaptic contacts, and F-actin flow. This pathway also influences NMDA receptor function through HRI-eIF2 alpha mediated translation, which is essential for excitatory synaptic transmission.

Wnt5a modulates dendritic spine dynamics through the regulation of Cofilin via small Rho GTPase activity in hippocampal neurons

Daniela Vallejo et al.

Summary: The study demonstrates that Wnt5a regulates the phosphorylation of Cofilin in neurons through Ras-related C3 botulinum toxin substrate 1 and cell division cycle 42, depending on subcellular compartment and extracellular calcium levels. Moreover, Wnt5a increases dendritic spine density and promotes maturation through Ras-related C3 botulinum toxin substrate 1, requiring the combined activation of small Rho GTPases to enhance filamentous actin levels and promote actin filament stability.
