Posttranslational regulation of mitochondrial frataxin and identification of compounds that increase frataxin levels in Friedreich's ataxia

Peter T. Hackett et al.

Summary: Friedreich's ataxia (FRDA) is a degenerative disease caused by a decrease in the mitochondrial protein frataxin (Fxn), leading to iron accumulation and impaired mitochondrial function. This study suggests that oxidant-mediated decreases in Fxn may contribute to FRDA progression, and some approved compounds have the potential to increase mitochondrial Fxn and delay disease progression.

Cur@SF NPs alleviate Friedreich's ataxia in a mouse model through synergistic iron chelation and antioxidation

Li Xu et al.

Summary: Friedrich's ataxia (FRDA) is a neurodegenerative disease characterized by abnormal iron metabolism, mitochondrial dysfunction, and oxidative damage. Curcumin nanoparticles (Cur@SF NPs) have shown therapeutic effects in improving FRDA symptoms by reducing oxidative stress and enhancing iron-sulfur cluster biogenesis.

Treatment with ROS detoxifying gold quantum clusters alleviates the functional decline in a mouse model of Friedreich ataxia

Chiara Villa et al.

Summary: FRDA is a disease caused by reduced expression of the mitochondrial protein frataxin due to a trinucleotide repeat expansion in the FXN gene. Research has shown that the use of gold cluster superstructures (Au-8-pXs) can improve mitochondrial function and alleviate FRDA-related pathology in cells. Systemic injection of Au-8-pXs also ameliorated motor function and cardiac contractility in a mouse model, suggesting potential therapeutic benefits for FRDA.
