Protein expression and secretion by filamentous fungi

Anup A. Sakekar et al.

Summary: Filamentous fungi are considered ideal microbial cell factories for protein expression and secretion. Even though our understanding of the secretory pathways in these organisms has improved significantly, their ability to produce foreign proteins still falls short of their potential. Further research into the molecular mechanisms of secretion pathways in filamentous fungi could lead to enhanced protein production, especially of valuable heterologous proteins.

Loss of EZH2-like or SU(VAR)3-9-like proteins causes simultaneous perturbations in H3K27 and H3K9 tri-methylation and associated developmental defects in the fungus Podospora anserina

F. Carlier et al.

Summary: Selective gene silencing is crucial for development, with H3K27me3-enriched heterochromatin maintaining transcriptional repression and regulating cell fate. Conversely, H3K9me3-enriched heterochromatin prevents differentiation and protects against transposable elements. Research using the fungus Podospora anserina revealed an interplay between these different heterochromatin conformations, with an interdependence of H3K9me3 and H3K27me3 marks observed. Loss of specific enzymes led to defects in developmental processes, highlighting the importance of histone modifications in fungal development.

Chromatin accessibility profiling in Neurospora crassa reveals molecular features associated with accessible and inaccessible chromatin

Aileen R. Ferraro et al.

Summary: Regulation of chromatin accessibility and transcription are coordinated processes. This study characterizes accessible chromatin regions in Neurospora crassa, revealing molecular features and histone modifications associated with accessibility and inaccessibility. H3K27 acetylation is the most predictive histone modification for open chromatin in N. crassa, while H3K36 methylation is a key marker of inaccessible chromatin.

Genome-wide functional screen of 3′UTR variants uncovers causal variants for human disease and evolution

Dustin Griesemer et al.

Summary: The study developed a method called MPRAu, which was used to sensitively assay over 12,000 3'UTR variants. It revealed the regulatory activity of 3'UTRs, mainly attributed to simple sequences, and uncovered diverse molecular mechanisms at base pair resolution. Additionally, the MPRAu identified hundreds of 3'UTR causal variants and potential metabolic evolutionary adaptations in East Asians.

High-resolution analysis of cell-state transitions in yeast suggests widespread transcriptional tuning by alternative starts

Minghao Chia et al.

Summary: An integrative analysis of cell-fate transitions in Saccharomyces cerevisiae reveals widespread elevation in alternative TSS and TES usage. The study highlights the regulatory role of alternative TSSs in gene expression through interactions with canonical promoters and distance between alternative and canonical TSSs. Specific chromatin modifiers play a crucial role in mediating gene repression via alternative TSSs, emphasizing the importance of upstream transcription in driving local changes in gene transcription.
