A toolkit for studying Varroa genomics and transcriptomics: preservation, extraction, and sequencing library preparation

Nonno Hasegawa et al.

Summary: This study developed methods for preserving Varroa DNA and RNA from the field to the lab and processing them into sequencing libraries. Preservation in ethanol successfully protected Varroa DNA and RNA, providing a simple and cost-effective method.

Direct Evidence for Infection of Varroa destructor Mites with the Bee-Pathogenic Deformed Wing Virus Variant B, but Not Variant A, via Fluorescence In Situ Hybridization Analysis

Sebastian Gisder et al.

Summary: Deformed wing virus (DWV), a bee-pathogenic virus, is spread by the mite Varroa destructor, leading to severe infections in bees. Different variants of DWV exist, and there is debate on whether the mite is a mechanical or biological vector of DWV.

Arthropods Under Pressure: Stress Responses and Immunity at the Pathogen-Vector Interface

Kristin L. Rosche et al.

Summary: Understanding the factors influencing the ability of arthropods to host and transmit pathogens is crucial for controlling vector-borne diseases. Arthropod immunity plays a central role in determining vector competence for pathogen acquisition and transmission, with immune responses triggered by microbial infections and stress on the host. Recent studies suggest a close connection between eukaryotic stress responses and innate immunity, with mechanisms such as the Unfolded Protein Response (UPR) and Integrated Stress Response (ISR) impacting immune signaling and potentially affecting vector competence in arthropods.

Adaptation to vector-based transmission in a honeybee virus

Amanda M. Norton et al.

Summary: Research shows that DWV genotypes A and B have different responses to mite numbers within honeybee colonies. DWV-A accumulation in honeybees is positively correlated with mite numbers, while DWV-B can persist with high loads in colonies even when mite numbers are low.

A New Strain of Virus Discovered in China Specific to the Parasitic Mite Varroa destructor Poses a Potential Threat to Honey Bees

Gongwen Chen et al.

Summary: A new strain of Varroa destructor virus-2 (VDV-2) was identified through metatranscriptome analysis of the microbial community associated with Varroa mites. An epidemiological investigation of the Chinese strain of VDV-2 (VDV-2 China) showed that the virus was highly prevalent among Varroa populations but was not found in adult worker bees from both Apis mellifera and Apis cerana colonies. The study also revealed a differential replication of the virus between the two host species, highlighting a potential threat to the Western honey bee population.

Single mosquito metatranscriptomics identifies vectors, emerging pathogens and reservoirs in one assay

Joshua Batson et al.

Summary: Unbiased metatranscriptomic sequencing of wild-caught mosquitoes can rapidly provide information on the various pathogens they carry, and analyze the host mosquito species, microbial prevalence, and viral co-infections. This method can provide valuable information for public health surveillance and intervention decisions.

Summary: The study focused on putative odorant carrier proteins in Varroa destructor and found that silencing of Vd40090 disrupted host selection, acceptance and feeding, as well as impaired gene expression related to reproduction in brood cells, leading to reduced reproduction and survival.
