Single-Vesicle Electrochemistry Reveals Sex Difference in Vesicular Storage and Release of Catecholamine

Summary: This study demonstrates the presence of sex differences in vesicle chemistry, specifically in the duration of exocytotic events and the extent of neurotransmitter release. Male rats show longer exocytotic events and release more neurotransmitter compared to female rats. Additionally, a higher percentage of vesicles in female rats exhibit partial release during exocytosis. These findings suggest that sex differences in the expression of calcium channels and membrane lipid composition may contribute to the observed differences in vesicle chemistry.

Concentration of stimulant regulates initial exocytotic molecular plasticity at single cells

Summary: This study examined the temporal and concentration effects of chemical stimulation on exocytotic molecular plasticity. The results showed that catecholamine release and release dynamics depended on the concentration of the stimulant solution. Increased release was observed at 30 and 50 mM stimulation, while decreased release was observed at 100 mM stimulation. These findings suggest a possible link between stimulation intensity and synaptic (or adrenal) plasticity.

Monitoring of Intracellular Vesicles in Cultured Neurons at Different Growth Stages Using Intracellular Vesicle Electrochemical Cytometry

Summary: By monitoring vesicular content and release kinetics in neurons at different growth stages using intracellular vesicle electrochemical cytometry, we observed an increase in neurotransmitter content and longer vesicle release process as neurons grew. Additionally, we found a shift in vesicular adsorption and rupture modes from dominant simple events to a coexistence of simple and complex events. This work may provide a new strategy for neuronal activity assessment and real-time cell activity analysis.

From FFN Dual Probe Screening to ITO Microdevice for Exocytosis Monitoring: Electrochemical and Fluorescence Requirements

Summary: In this study, four new fluorescent false neurotransmitters (FFN) probes were synthesized and their spectroscopic and electrochemical properties were investigated. The results showed that one of the probes (FFN122) had the optimal suitability for fluorescence microscopy. However, it was found that this probe did not accumulate into secretory vesicles of model cells. Therefore, FFN42 is currently the best bioelectrofluorescent compromise. Additionally, the effects of a transparent and conducting indium tin oxide (ITO) microdevice on electrochemical performances and reusability were examined.

Sensitive detection of electrophysiology and dopamine vesicular exocytosis of hESC-derived dopaminergic neurons using multifunctional microelectrode array

Summary: A multifunctional microelectrode array was developed to detect electrophysiological signals and dopamine vesicular exocytosis of human embryonic stem cells derived dopaminergic neurons (hESC-DDNs). The modified microelectrode array demonstrated low noise, high signal-to-noise ratio, high sensitivity and high time resolution for detecting weak current signals generated by dopamine vesicular exocytosis. The study also revealed an important mode of dopamine vesicular exocytosis in hESC-DDNs.
