Interest of novel N-alkylpyridinium-indolizine hybrids in the field of Alzheimer's disease: Synthesis, characterization and evaluation of antioxidant activity, cholinesterase inhibition, and amyloid fibrillation interference

Isabelle Baussanne et al.

Summary: A small library of molecules combining indolizine and N-alkyl pyridinium was synthesized and evaluated for Alzheimer's disease treatment. The new compounds were classified in three series based on the number of methylene residues linking the two heterocycles, and their activities against AD targets were assessed in vitro.

Sodium fusidate prevents protein aggregation of silk fibroin and offers new perspectives for human lens material disaggregation

Oktay K. Gasymov et al.

Summary: Silk fibroin is prone to forming amyloid-like aggregated species in solution, but fusidic acid acts as an anti-aggregating agent for silk fibroin by inducing changes in the environment of its aromatic residues. Fusidic acid, already approved for human use as a drug and used in ophthalmic preparations, has potential as a therapeutic treatment for cataract and other protein aggregation disorders.

Curcumin ligand based iridium(III) complexes as inhibition and visualization agent of beta-amyloid fibrillation

Yunwei Zhao et al.

Summary: In this work, a series of curcumin ligand based Ir(III) complexes were synthesized and investigated for their outstanding ability to inhibit and monitor the fibrillation of A beta 42 peptides. The experimental results showed that these complexes can effectively inhibit and monitor A beta 42 peptide aggregation in vitro, with some complexes even completely inhibiting fibrillation at a concentration of 50 mu M. Additionally, these complexes demonstrated low cytotoxicity and neuro-protective ability against the cytotoxicity caused by three forms of A beta 42 peptides, showing potential for therapeutic applications in Alzheimer's disease.

Caffeic acid for the prevention and treatment of Alzheimer's disease: The effect of lipid membranes on the inhibition of aggregation and disruption of Aβ fibrils

Stephanie Andrade et al.

Summary: The study found that caffeic acid (CA) exhibited strong activity in preventing the aggregation and disaggregation of A beta 1-42, but its disaggregating activity increased over time in the presence of lipid membranes. This suggests the therapeutic potential of CA in preventing and treating Alzheimer's disease, warranting further investigations in animal models.

Inhibition behavior of Sennoside A and Sennoside C on amyloid fibrillation of human lysozyme and its possible mechanism

Wen Gao et al.

Summary: In this study, the inhibitory effects of Sennoside A and Sennoside C on amyloid fibrillation were evaluated using human lysozyme as a model. Both compounds were found to inhibit the fibrillation of lysozyme in a dose-dependent manner, with interactions mainly through hydrogen bonds, vander-Waals forces, and hydrophobic interactions. Transmission electron microscopy and atomic force microscopy confirmed the reduction in fibrillation, supporting the conclusion that both Sennoside A and Sennoside C can significantly inhibit amyloid fibrillation of lysozyme.

Binding to Amyloid-β Protein by Photothermal Blood-Brain Barrier-Penetrating Nanoparticles for Inhibition and Disaggregation of Fibrillation

Hao Geng et al.

Summary: The excess accumulation of amyloid-beta (A beta) protein in the brain is the primary cause of Alzheimer's disease (AD). Inhibition of A beta fibrillation and disaggregation of A beta fibrils using near infrared (NIR) light-responsive nanoparticles (NPs) shows promising therapeutic potential for AD.
