Substrate-Inspired Fragment Merging and Growing Affords Efficacious LasB Inhibitors

Summary: Extracellular virulence factors, including the LasB enzyme secreted by the Gram-negative pathogen Pseudomonas aeruginosa, have become important targets in the current antimicrobial resistance crisis. In this study, a sub-micromolar, non-peptidic inhibitor of LasB was discovered through structural data analysis and optimization, showing improved in vivo efficacy in Galleria mellonella larvae. This class of compounds shows promise as potential drug candidates for combating infections.

Structure-Based Design of α-Substituted Mercaptoacetamides as Inhibitors of the Virulence Factor LasB from Pseudomonas aeruginosa

Summary: Antivirulence therapy is an effective method to combat infections caused by multidrug-resistant bacteria. In this study, we explored a novel class of inhibitors that target elastase, a crucial virulence factor in Pseudomonas aeruginosa. These inhibitors showed maintained inhibitory activity, selectivity, and increased hydrophilicity. They were found to reduce bacterial pathogenicity and maintain the integrity of lung and skin cells in the diseased state. Moreover, two promising compounds improved the survival rate of Galleria mellonella larvae treated with P. aeruginosa culture supernatant.
