Summary: A novel series of homonucleosides and their double-headed analogs were designed and synthesized, characterized by various spectroscopic techniques, and screened for cytotoxic activities against four cell lines. The double-headed analogue 18 showed significant growth inhibition in all cell lines tested, inducing apoptosis through caspase-3/7 activation and cell-cycle arrest. Molecular docking studies confirmed that compound 18 activated caspase-3 via hydrogen bonding and hydrophobic interactions.

Identification of NSAIDs as lipoxygenase inhibitors through highly sensitive chemiluminescence method, expression analysis in mononuclear cells and computational studies

Summary: The inhibitory effects of nine non-steroidal anti-inflammatory drugs (NSAIDs) on soybean 15-lipoxygenase (15-LOX) enzyme were investigated using UV-absorbance, colorimetric, and chemiluminescence methods. Only Ibuprofen and Ketoprofen exhibited enzyme inhibition via UV-absorbance, while the chemiluminescence method proved to be the most sensitive for identifying 15-LOX inhibitors. The IC50 values of the drugs ranged from 3.52 +/- 0.08 to 62.6 +/- 2.15 mu M, with Naproxen showing the strongest inhibition and Piroxicam and Tenoxicam being the least active inhibitors.

Investigating the effects of two novel 4-MMPB analogs as potent lipoxygenase inhibitors for prostate cancer treatment

Summary: 4-PMPB and 4-EMPB exhibit similar anti-cancer effects on PC-3 cells compared to 4-MMPB, while being ineffective on normal cells. Apoptosis and ferroptosis were identified as the main mechanisms of induced cell death in these cancerous cells. In vivo results showed that both analogs had similar anti-cancer effects as 4-MMPB, leading to delayed tumor growth without noticeable side effects in weight loss and histological investigations.

Summary: A class of structurally unique para-aminobenzenesulfonyl oxadiazoles were designed and synthesized as potential antimicrobial agents, with notable antibacterial potency exhibited by hexyl derivative 8b. Compound 8b could effectively disrupt the membrane and form a steady 8b-DNA complex to potentially cause bacterial metabolic inactivation. Molecular docking revealed that 8b could interact with DNA topoisomerase IV via noncovalent interactions to hinder enzyme function. Hexyl derivative 8b shows promise as a new lead compound for further investigation.
