Microwave-assisted N-alkylation of amines with alcohols catalyzed by MnCl2: Anticancer, docking, and DFT studies

Summary: A new protocol for the synthesis of tertiary amines through the N-alkylation of amines with alcohols using MnCl2 as a catalyst under microwave conditions is described. The synthesized compounds were tested for anticancer efficacy and compound 3h demonstrated potent activity against multiple cancer cell lines, surpassing the reference drug cisplatin. The introduction of an electron-withdrawing group on the phenyl ring resulted in increased anticancer activity. Docking analysis and in vitro assessment revealed high affinity of the compounds towards HER2 kinase, indicating their potential as promising anticancer agents.

Design, synthesis and molecular docking studies of imidazole and benzimidazole linked ethionamide derivatives as inhibitors of InhA and antituberculosis agents

Summary: A new class of compounds consisting of imidazole and benzimidazole analogs linked to ethionamide were designed and synthesized to explore effective antituberculosis agents. Both experimental and computational methods were employed to evaluate their antimicrobial activity against Mycobacterium tuberculosis, as well as their drug likeness and ADMET properties. The compounds exhibited high potency and low cytotoxicity, making them attractive candidates for the development of new and improved anti-TB drugs.

Synthesis, in silico ADME, toxicity prediction and molecular docking studies of N-substituted [1,2,4]triazolo[4,3-a]pyrazine derivatives as potential anticonvulsant agents

Summary: Two new series of pyrazine derivatives were synthesized and evaluated for their antioxidant, anticonvulsant, and neurotoxicity properties. Compounds 2a, 2b, and 2e showed good anticonvulsant activity and minimal neurotoxicity. Computational analysis revealed strong binding interactions of these compounds in the binding pocket of a specific protein. The results suggest that compounds 2a, 2b, and 2e have potential as safer and more effective anticonvulsant drugs with neuroprotective properties.

Design and synthesis of novel benzimidazole linked thiazole derivatives as promising inhibitors of drug-resistant tuberculosis

Summary: This study successfully developed a new class of benzimidazole-linked thiazole compounds as chemotherapeutic drugs with anti-tuberculosis activity. Some of these compounds showed high antimicrobial effects against drug-resistant Mycobacterium tuberculosis strains. The study also revealed some structure-activity relationships and demonstrated the interaction of the compounds with the key protein MmpL3 through molecular docking studies. In addition, the potential drug properties of these compounds were validated through in-silico predictions.
