Results of the phase I CCTG IND.231 trial of CX-5461 in patients with advanced solid tumors enriched for DNA-repair deficiencies

John Hilton et al.

Summary: CX-5461, a G-quadruplex stabilizer, exhibits synthetic lethality in homologous recombination-deficient models. This multicentre phase I trial in patients with solid tumors demonstrates the clinical proof-of-concept for CX-5461, with a recommended phase II dose of 475 mg/m(2) in patients with defective homologous recombination. Responses are observed in 14% of patients, primarily in those with defective homologous recombination.

The i-Motif as a Molecular Target: More Than a Complementary DNA Secondary Structure

Susie L. Brown et al.

Summary: The i-motif is a dynamic secondary DNA structure rich in cytosine, with potential as a molecular switch for gene expression control. It may play a role in facilitating or hindering expansion of DNA elements in genomic areas with repetitive nucleotide sequences. Recent discoveries, including the identification of a specific i-motif antibody, are advancing the field of i-motif research.

The chemotherapeutic CX-5461 primarily targets TOP2B and exhibits selective activity in high-risk neuroblastoma

Min Pan et al.

Summary: The study found that the small molecule CX-5461 has selective cytotoxicity in high-risk neuroblastoma and synergizes with topoisomerase I inhibitors to improve survival. However, it was discovered that the primary target of CX-5461 in this disease is topoisomerase II beta, not RNA-POL I, raising unexpected safety concerns.

Transcription-associated topoisomerase 2α a (TOP2A) activity is a major effector of cytotoxicity induced by G-quadruplex ligands

Madeleine Bossaert et al.

Summary: Through an unbiased genetic approach, the study identified TOP2A as a major effector of cytotoxicity induced by G4 ligands. The study shows that both TOP2 activity and transcription play crucial roles in DNA break production following G4 ligand treatments. In contrast, the clastogenic activity of G4 ligands is counteracted by TOP1 and factors promoting transcriptional elongation.
