Edaravone ameliorates depressive and anxiety-like behaviors via Sirt1/Nrf2/HO-1/Gpx4 pathway

Summary: Inflammation and oxidative stress have been implicated in the development of depression. This study investigated the antidepressant effects and mechanisms of action of Edaravone (EDA), a free radical scavenger. The results demonstrated that EDA alleviated depressive and anxiety-like behaviors in a chronic social defeat stress (CSDS) depression model. Furthermore, EDA reduced neuronal loss, oxidative stress damage, and activation of inflammatory cytokines in the hippocampus and medial prefrontal cortex. The antidepressant effects of EDA were mediated through the Sirt1/Nrf2/HO-1/Gpx4 signaling pathway and Gpx4-mediated ferroptosis.

Summary: In patients with age-related macular degeneration (AMD), there are structural and functional defects in crucial retinal pigment epithelial (RPE) cells' mitochondria, along with deficient expression of Dicer enzyme. This deficiency increases the expression of Alu RNA, damaging mitochondria and inducing cell apoptosis, thereby driving the progression of dry AMD. It is proposed that up-regulating mitophagy and mitochondrial biogenesis (MB) can mitigate the adverse effects of Alu RNA on RPE cells, and activating Sirt1, AMPK, Nrf2, and PPAR alpha may be useful in this regard.
