GPNMB confers risk for Parkinson's disease through interaction with α-synuclein

Maria E. Diaz-Ortiz et al.

Summary: Through genome-wide association studies, a risk gene called GPNMB has been identified for Parkinson's disease, and its interaction and function with α-synuclein have been discovered. In PD patients, elevated levels of GPNMB in plasma are associated with disease severity.

Antihypertensive Medication Classes and the Risk of Dementia: A Systematic Review and Network Meta-Analysis

Melina G. H. E. den Brok et al.

Summary: This study found that in observational studies, treatment with calcium channel blockers (CCBs) or angiotensin II receptor blockers (ARBs) was associated with lower dementia risks. However, there was insufficient randomized controlled trials (RCTs) data to create a robust network based on randomized data alone.

Genetic control of the human brain proteome

Chloe Robins et al.

Summary: The study generated an online brain pQTL resource for 7,376 proteins by analyzing genetic and proteomic data from the dorsolateral prefrontal cortex of 330 older adults. The identified pQTLs tend to be non-synonymous variations, over-represented among brain disease-associated variants, and show good replication in an independent brain dataset. Comparison with brain eQTLs revealed that most pQTLs are also eQTLs, suggesting lower pQTL mapping power and greater evolutionary constraint on protein abundance.

Common and rare variant association analyses in amyotrophic lateral sclerosis identify 15 risk loci with distinct genetic architectures and neuron-specific biology

Wouter van Rheenen et al.

Summary: A cross-ancestry genome-wide association meta-analysis of amyotrophic lateral sclerosis (ALS) identified 15 risk loci with distinct genetic architectures and neuron-specific biology. The study emphasized the urgent need for disease-modifying therapies for ALS. High cholesterol levels were suggested to have a causal role in the onset of ALS, among other environmental and lifestyle risk factors.

Integrating human brain proteomes with genome-wide association data implicates new proteins in Alzheimer's disease pathogenesis

Aliza P. Wingo et al.

Summary: This study identified 11 genes that may play a role in Alzheimer's disease (AD) by affecting brain protein abundance, through a proteome-wide association study. Eight of these genes are new AD risk genes not identified before by AD GWAS, providing new insights into AD pathogenesis. This research offers promising targets for further mechanistic and therapeutic studies on AD.

Genome-wide meta-analysis, fine-mapping and integrative prioritization implicate new Alzheimer's disease risk genes

Jeremy Schwartzentruber et al.

Summary: A genome-wide AD meta-analysis identified 37 risk loci, including new associations near CCDC6, TSPAN14, NCK2, and SPRED2. By fine-mapping SNPs and using functional annotation, 21 SNPs with >50% probability of being causally involved in AD risk were identified. Protein interaction networks and tissue-specific expression helped prioritize genes, leading to the discovery of likely causal genes.

Genome sequencing analysis identifies new loci associated with Lewy body dementia and provides insights into its genetic architecture

Ruth Chia et al.

Summary: By conducting whole-genome sequencing and genetic analysis, researchers have identified common genetic risk factors and pathways shared between Lewy body dementia, Alzheimer's disease, and Parkinson's disease. This provides a deeper understanding of the complex genetic architecture of these age-related neurodegenerative conditions.

A Neanderthal OAS1 isoform protects individuals of European ancestry against COVID-19 susceptibility and severity

Sirui Zhou et al.

Summary: The study identified that increased levels of OAS1 protein are associated with reduced susceptibility and severity of COVID-19, providing evidence for a protective role of OAS1 in adverse outcomes. This suggests that pharmacological agents that increase OAS1 levels could be prioritized for drug development against COVID-19. The variant of the OAS1 gene associated with decreased risk of death in COVID-19 patients encodes an enzyme critical for the innate immune response to viral infections.

Identification of Candidate Parkinson Disease Genes by Integrating Genome-Wide Association Study, Expression, and Epigenetic Data Sets

Demis A. Kia et al.

Summary: This study utilized bioinformatic tools Coloc and TWAS to analyze PD GWAS data from 2017, identifying gene expression and splicing mechanisms associated with PD risk, further characterized using cell-type specificity and weighted coexpression networks. Results suggest that several candidate genes and pathways are associated with the findings observed in PD GWAS studies.

Genomic atlas of the proteome from brain, CSF and plasma prioritizes proteins implicated in neurological disorders

Chengran Yang et al.

Summary: Yang et al. created a genomic atlas of protein levels in brain, cerebrospinal fluid, and plasma, identifying proteins linked to neurological diseases and potential drug targets. They found tissue-specific differences in protein quantitative trait loci (pQTLs) and observed that some pQTLs did not co-localize with other genetic regulation markers. This multi-tissue study could help map signals from genome-wide association studies to functional genes and discover drug targets for neurological diseases.
