Electrophysiological Properties of Induced Pluripotent Stem Cell-Derived Midbrain Dopaminergic Neurons Correlate With Expression of Tyrosine Hydroxylase

Aleksandar Rakovic et al.

Summary: Induced pluripotent stem cells can be used to create patient-specific in vitro models for studying Parkinson's disease and drug screening. However, current differentiation methods result in heterogeneous cell populations with low fractions of relevant dopaminergic neurons. This study utilized CRISPR/Cas9 technology to generate a reporter iPSC line and compared different differentiation protocols. The floor plate protocol was found to be the most efficient in generating electrophysiologically mature dopaminergic neurons.

Preclinical Development of 18F-OF-NB1 for Imaging GluN2B-Containing N-Methyl-D-Aspartate Receptors and Its Utility as a Biomarker for Amyotrophic Lateral Sclerosis

Hazem Ahmed et al.

Summary: The study successfully developed a highly efficient PET probe, F-18-OF-NB1, for imaging the GluN2B subunits of the N-methyl-D-aspartate receptor. The probe has the potential for application in PET imaging studies in ALS patients and other brain disorders.

Cell-Type-Specific High Throughput Toxicity Testing in Human Midbrain Organoids

Henrik Renner et al.

Summary: Toxicity testing is essential in the development and approval of chemical compounds, and 3D human organoid systems show promise in providing more relevant models of human physiology. Recent advancements in standardized automated midbrain organoids have allowed for efficient evaluation of toxic effects, with high reproducibility. This study demonstrates the feasibility of quantitatively assessing cell-type-specific toxicity in human organoids in vitro, paving the way for more accurate toxicity predictions.
