Insights into animal septins using recombinant human septin octamers with distinct SEPT9 isoforms

Summary: This study successfully isolated recombinant human septin octamers containing different SEPT9 isoforms for the first time, confirming their octameric nature in solution. Reconstitution studies showed that octamers with either long or short SEPT9 isoforms can form filament assemblies and directly bind and cross-link actin filaments, suggesting that septin-decorated actin structures in cells may reflect direct actin-septin interactions. These recombinant SEPT9-containing octamers will enable the design of cell-free assays to study the complex interactions of septins with cell membranes and the actin and microtubule cytoskeleton.

An atomic model for the human septin hexamer by cryo-EM

Summary: Human septins must form hetero-oligomeric rod-like particles to create functional filaments, and a cryo-electron microscopy structure has revealed the internal cavity and interactions between component subunits within the hexameric rod. The elongated particle exhibits bending at the center where SEPT7 forms a homodimeric interface, which is likely related to the ability of septin filaments to recognize and induce membrane curvature.
