Phosphomimetic Mutation at Ser165 of α-Tubulin Promotes the Persistence of GTP Caps in Microtubules

Summary: This study found that protein kinase C-mediated phosphorylation of alpha-tubulin at a specific site can stimulate microtubule polymerization. Molecular simulations revealed that the phosphomimetic variant S165D alters the spatial relationship between alpha:Glu254 and beta:GTP, thereby slowing GTP hydrolysis and promoting the formation of GTP caps. Cellular experiments confirmed these findings.

GTP-dependent formation of straight tubulin oligomers leads to microtubule nucleation

Summary: This study demonstrates that the formation of straight oligomers of critical size is essential for nucleation of microtubules. Both GDP and GTP tubulin form single-stranded oligomers with different curvature distributions, and the proportion of straight oligomers can accelerate nucleation. The results suggest that cellular factors involved in nucleation promote it via stabilization of straight oligomers.

Hierarchical Assembly Pathways of Spermine-Induced Tubulin Conical-Spiral Architectures

Summary: This study demonstrates that spermine can induce the in vitro assembly of tubulin into various hierarchical structures, including conical-spiral tubes and inverted hexagonal bundles, depending on the concentration of spermine. The increase in spermine concentration leads to longer tubes and the formation of a new type of tubulin assembly. These findings provide insights into the versatile morphologies of tubulin assemblies and the potential interactions controlling protein-based biomaterial composition and construction.
