Methylotrophy of Pichia pastoris: Current Advances, Applications, and Future Perspectives for Methanol-Based Biomanufacturing

The article discusses the current understanding of methanol metabolism in P. pastoris, important factors influencing its methanol utilization ability, recent advances in using engineered P. pastoris for producing various chemicals from methanol, as well as future challenges and potential solutions in developing P. pastoris as an efficient cell factory for methanol-based biomanufacturing.

Methanol oxidase from Hansenula polymorpha shows activity in peroxisome-deficient Pichia pastoris

Summary: In this study, a genetically engineered strain of Pichia pastoris with peroxisome defects, Apex3, was constructed to simulate the damage caused by high concentrations of methanol. By introducing methanol oxidase (MOX) from Hansenula polymorpha, a new recombinant strain Apex3-MOX was obtained, showing improved alcohol oxidase activity and higher biomass compared to Apex3. The ability to utilize and tolerate higher methanol concentrations was partially enhanced by the introduction of MOX.
