Development of Integrated Approaches to Testing and Assessment (IATA) case studies on developmental neurotoxicity (DNT) risk assessment

Summary: The EFSA Panel on Plant Protection Products and their Residues conducted two AOP-informed IATA case studies to address the DNT hazard identification and characterisation problem for pesticide active substances deltamethrin and flufenacet. Through systematic literature review, expert knowledge elicitation, and probabilistic approach, the studies explored the applicability of the DNT in vitro testing battery in regulatory risk assessment. This evidence-based approach allowed for conclusions with an acceptable level of certainty in DNT hazard identification and characterisation.

Toward a Better Testing Paradigm for Developmental Neurotoxicity: OECD Efforts and Regulatory Considerations

Summary: Recognizing the current DNT testing paradigm is not suitable for assessing a large number of chemicals, scientific advances have been made in alternative methods to evaluate chemical interactions with the developing nervous system without using animals. The OECD provides internationally harmonised guidelines based on cellular models for testing and assessing chemicals for DNT. To address challenging endpoints like DNT, the OECD has been developing integrated approaches for testing and assessment, which rely on a combination of different data layers and mechanistic knowledge.
