Streptomyces coelicolor Vesicles: Many Molecules To Be Delivered

Teresa Faddetta et al.

Summary: This study investigated the production and characterization of membrane vesicles (MVs) in Streptomyces coelicolor M145. The MVs were found to contain various proteins and metabolites, including those involved in cell metabolism/differentiation, molecular processing/transport, and stress response. Additionally, the MVs contained a range of metabolites, such as antibiotics, vitamins, amino acids, and components of carbon metabolism. These findings provide valuable insights into the biogenesis and functions of MVs.

Simultaneous Raman and infrared spectroscopy: a novel combination for studying bacterial infections at the single cell level

Cassio Lima et al.

Summary: Sepsis is a life-threatening condition that causes approximately 11 million deaths worldwide. Raman and infrared spectroscopy, as diagnostic tools, have the potential to rapidly detect bacterial infections. This study introduces a new technology that combines infrared and Raman spectra, and demonstrates its similarity and reproducibility in analyzing bacterial samples. The findings show different clustering patterns between the two methods, highlighting the complementary information provided by microbial pigments in Raman spectra. Overall, the combination of Raman and infrared spectroscopy has promising applications in clinical microbiology.

Single-cell metabolite detection and genomics reveals uncultivated talented producer

Masato Kogawa et al.

Summary: This study presents an analysis pipeline for the efficient identification of uncultivated producers in the microbiomes, highlighting the importance of complementary technologies. It reveals a new bacterium as the producer of previously unassigned antifungal compounds in a chemically rich bacteriosponge, suggesting an even higher chemical richness than previously appreciated. The research also shows remarkable parallels between distantly related symbionts of the same host and adds functional support for diverse chemically prolific lineages in microbial dark matter.

Deconstruction of Obscure Features in SVD-Decomposed Raman Images from P. chrysogenum Reveals Complex Mixing of Spectra from Five Cellular Constituents

Ashok Zachariah Samuel et al.

Summary: Raman imaging has evolved into a molecular profiling technique in biomedical applications, with SVD and PCA commonly used for data analysis. However, these methods often fail to fully interpret the molecular information in the original data. By developing a combined multivariate approach, this study successfully extracted the contribution of five biomolecular constituents of Penicillium chrysogenum to the SVD vectors, advancing SVD/PCA into a more meaningful molecular domain.

Machine learning-assisted single-cell Raman fingerprinting for in situ and nondestructive classification of prokaryotes

Nanako Kanno et al.

Summary: Utilizing a combination of machine learning and single-cell Raman microspectroscopy allows for accurate classification of phylogenetically diverse prokaryotes, shedding light on microbial dark matter. The presence of carotenoids and membrane lipid structure play key roles in distinguishing prokaryotic species, with unique Raman markers identified for an ammonia-oxidizing archaeon.
