Heterologous expression and characterization of two novel glucanases derived from sheep rumen microbiota

A novel bifunctional glucanase exhibiting high production of glucose and cellobiose from rumen bacterium

Summary: In this study, two novel glucanase genes, Cel5A-h38 and Cel5A-h49, were isolated and heterologously expressed from the metatranscriptome of the Hu sheep rumen microbiome. The enzymes displayed distinct substrate specificity, with Cel5A-h38 being active exclusively in the presence of mixed-linked glucans and Cel5A-h49 exhibiting a wider substrate spectrum. The results provided new insights into glucanase activity and established a foundation for the bioconversion of lignocellulosic biomass.

Rational design and structure-based engineering of alkaline pectate lyase from Paenibacillus sp. 0602 to improve thermostability

Summary: In this study, the thermostability of an alkaline pectate lyase was improved through rational design and structure-based engineering. Mutants G241A and G241V exhibited higher thermostability compared to the wild-type, and the combinational mutant K93I/G241A showed simultaneous improvements in thermostability and specific activity. The results suggest that the engineered enzyme is more suitable for industrial production and the strategies used in this study could be applied to other industrial enzymes.
