Voxel-based meta-analysis of gray matter abnormalities in idiopathic dystonia

Summary: In this study, a meta-analysis was conducted to investigate gray matter changes in patients with idiopathic dystonia. The results showed consistent widespread gray matter abnormalities in various brain regions. These abnormalities were shared among different subtypes of idiopathic dystonia and were not limited to the corticostriatal circuits.

Short- and Long-term Central Action of Botulinum Neurotoxin Treatment in Laryngeal Dystonia

Summary: This study investigated the short-term and long-term effects of BoNT treatment on brain function in patients with LD. The results suggest that BoNT-treated patients who benefited from the treatment had reduced activity in the left precuneus compared to untreated patients and patients who did not benefit from the treatment. Additionally, BoNT-treated patients with adductor LD showed decreased activity in the right thalamus, while patients with abductor LD showed reduced activity in the left inferior frontal cortex. There were no significant differences in brain activity between patients with shorter and longer treatment durations. However, patients with intermediate treatment duration of 6-12 years showed reduced activity in the right cerebellum and right prefrontal cortex compared to patients with shorter treatment duration.

Neural endophenotypes and predictors of laryngeal dystonia penetrance and manifestation

Summary: The study identified shared neural endophenotypes between patients with laryngeal dystonia and their unaffected relatives, representing the endophenotypic marker of dystonia penetrance. Additional abnormalities found only in patients likely represent a secondary endophenotype of dystonia manifestation. Machine learning was able to predict the risk for dystonia development in susceptible individuals based on neural features, showing potential for screening at-risk individuals and selecting families for genetic studies or clinical trials.
