Synthesis of Chiral Hydantoins and Thiazolidinediones via Iridium-Catalyzed Asymmetric Hydrogenation

Summary: In this work, we developed an iridium-catalyzed asymmetric hydrogenation of hydantoin and thiazolidinedione derived exocyclic alkenes using BiphPHOX as a ligand. The reaction showed excellent functional group tolerance and provided the hydrogenated products with high yields and enantioselectivities. Moreover, the hydrogenated product can be efficiently transformed into an intermediate for the synthesis of a HIV protease inhibitor.

Despite the pH neutrality of these precatalysts, the reaction mixtures become acidic under hydrogenation conditions. The paper explores the implications of this acidity on asymmetric hydrogenation and suggests opportunities for utilizing or eliminating the generated Bronsted acid.

Combined Theoretical and Experimental Studies Unravel Multiple Pathways to Convergent Asymmetric Hydrogenation of Enamides

Summary: A highly efficient convergent asymmetric hydrogenation of E/Z mixtures of enamides catalyzed by N,P-iridium complexes is presented in this study. Mechanistic studies revealed different pathways for enantioconvergence, including fast isomerization for aryl enamides and substrate chelation for alkyl enamides. DFT calculations were performed to predict the correct absolute configuration of the products and strengthen the proposed mechanism of the iridium-catalyzed isomerization pathway.

Summary: A family of phosphite/phosphinite-thioether ligands has shown excellent catalytic performance in the Ir-catalyzed asymmetric hydrogenation of a variety of olefins. Their modular architecture allows for a broad substrate scope, covering different substitution patterns and functional groups. The catalytic performance of the Ir-ligand assemblies remains consistent when using environmentally friendly 1,2-propylene carbonate as a solvent.

Synthesis of Chiral 2-Substituted 1,4-Benzoxazin-3-ones via Iridium-Catalyzed Enantioselective Hydrogenation of Benzoxazinones

Summary: An efficient iridium-catalyzed enantioselective hydrogenation method using iPr-BiphPHOX ligand for 2-alkylidene 1,4-benzoxazin-3-ones was reported, delivering reduced products with excellent yields (up to 99%) and enantioselectivities (up to 99% ee). The reaction worked well on a gram scale with low catalyst loadings and no erosion in enantioselectivity. Additionally, three bioactive molecules can be easily obtained from the reduced products.

Gram-scale asymmetric hydrogenation proceeded well with low-catalyst loading, and a possible reaction process was revealed based on various experiments conducted.
