Sheathless acoustic based flow cell sorter for enrichment of rare cells

Summary: Cell enrichment is a powerful tool in various cell research applications, especially for low abundance cell types. A microfluidic fluorescence activated cell sorting device was developed to detect and sort cells with high efficiency. The device was able to remove over 98% of white blood cells and recover labeled lymphocytes with 80% efficiency, as well as successfully isolating fetal nucleated red blood cells from clinical samples for genetic analysis, showing potential for biomedical applications involving fetal cells, circulating tumor cells, and stem cells.

Summary: Immune response dysregulation plays a key role in the pathogenesis of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). The study evaluated immune and endothelial blood cell profiles of COVID-19 patients, revealing depletion of T cell subsets and an increase in CD56(+) monocytes in severe cases. Age, obesity, and hypertension were found to synergistically impact immune markers and disease severity in SARS-CoV-2 infection.

Summary: This study aimed to explore the feasibility of using NanoVelcro Chips to simultaneously detect single CTCs and CTC clusters in neuroendocrine tumors (NETs), as well as to investigate their potential role as biomarkers for monitoring patient response after peptide receptor radionuclide therapy (PRRT). Preliminary findings suggest that both CTC clusters and single CTCs may offer a potential noninvasive option for monitoring treatment response in NET patients undergoing PRRT.

Confined placental mosaicism and the association with pregnancy outcome and fetal growth: a review of the literature

Summary: The review evaluated the association between confined placental mosaicism (CPM) and prenatal growth and adverse pregnancy outcomes. Evidence showed that CPM was significantly associated with low birth weight, fetal growth restriction (FGR), and a high rate of preterm birth and structural fetal anomalies. Higher levels of mosaicism in chorionic villus sampling and the presence of uniparental disomy were also linked to adverse pregnancy outcomes. Monitoring fetal growth, examining for anomalies, and raising awareness of potential premature birth are advised for clinicians when CPM is detected.

Smart-RRBS for single-cell methylome and transcriptome analysis

Summary: The integration of DNA methylation and transcriptional state within single cells is of broad interest for studying cellular heterogeneity and rare cell populations. The Smart-RRBS protocol combines Smart-seq2 and RRBS to generate paired epigenetic promoter and RNA-expression measurements for approximately 24% of protein-coding genes in typical single cells, and can also be applied to tissue samples comprising hundreds of cells. The protocol, excluding flow sorting of cells and sequencing, takes around 3 days to process up to 192 samples manually and requires basic molecular biology expertise and laboratory equipment.

Circulating trophoblast cell clusters for early detection of placenta accreta spectrum disorders

Summary: A newly developed microchip technology enables efficient enrichment of circulating trophoblasts from maternal blood for early detection of placenta accreta spectrum (PAS). The researchers identified a high prevalence of circulating trophoblast clusters in PAS patients and constructed a diagnostic model with excellent performance in distinguishing PAS. The combined trophoblast assay shows potential to improve current diagnostic methods for the early detection of PAS.
