Discrimination of isomeric trisaccharides and their relative quantification in honeys using trapped ion mobility spectrometry

By using electrospray ionization coupled to trapped ion mobility spectrometry, it is possible to discriminate multiple isomeric trisaccharides and determine the origin of honeys based on their relative content and ratios. This direct approach provides an alternative to longer chromatographic runs for routine analysis.

Anomeric Retention of Carbohydrates in Multistage Cyclic Ion Mobility (IMSn): De Novo Structural Elucidation of Enzymatically Produced Mannosides

Summary: Carbohydrates are complex structures with different levels of isomerism, notably the anomerism of the glycosidic bond, which are still challenging analysts today. Recent research has shown that anomerism can be preserved upon gas-phase fragmentation and high-resolution ion mobility (IMS) can distinguish anomers. The study demonstrates that high-resolution IMS and IMSn can be used to characterize the intrachain anomerism in tri- and tetrasaccharides in a biological medium, providing the first evidence that a single carbohydrate-active enzyme can synthesize both alpha- and beta-glycosidic linkages.

However, traditional IR spectroscopy requires laser systems and long measurement times, limiting its utility. A new high-throughput instrument combining ion-mobility spectrometry and cryogenic IR spectroscopy has been developed, allowing faster acquisition of IR fingerprints for isomeric glycans.
