Internal calibration as an emerging approach for endogenous analyte quantification: Application to steroids

Summary: In this study, an innovative internal calibration approach using stable isotope-labeled standard version of endogenous compound was developed to improve the quantitative performance of mass spectrometry methods. The results showed that the response factors obtained on different LC-MS platforms were consistent and the quantitative results in human serum samples were similar to those obtained with conventional quantification approach based on external calibration. Furthermore, it was demonstrated that the naturally occurring isotopes of SIL could be used to increase the accuracy for analytes with low concentrations.

Advancing D-amino acid-containing peptide discovery in the metazoan

Summary: The discovery of enzyme-derived D-amino acid-containing peptides challenges assumptions about the homochirality of animal proteins and presents new analytical challenges in peptide structural and functional characterization. Most known DAACPs have been identified through laborious purification studies or homology to previously identified ones. High throughput mass spectrometry-based peptidomics dominate peptide characterization experiments, with stereochemistry rarely considered due to technical challenges of identifying L/D isomerization, suggesting that more DAACPs remain to be uncovered.

Analysis of Peptide Stereochemistry in Single Cells by Capillary Electrophoresis-Trapped Ion Mobility Spectrometry Mass Spectrometry

Summary: Single cell analysis investigates molecular heterogeneity in morphologically similar cell populations through quantitative or qualitative measurements of genetic, proteomic, or metabolic products. This study presents a powerful method for analyzing PIPx profiles in biological samples using a chiral polysaccharide stationary phase and high-resolution mass spectrometry.
