Exploiting the orthogonal CRISPR-Cas12a/Cas13a trans-cleavage for dual-gene virus detection using a handheld device

Tian Tian et al.

Summary: This study introduces a highly efficient dual-gene detection technique based on the orthogonal DNA/RNA collateral cleavage mechanism of the Cas12a/Cas13a system. The technique allows simultaneous detection of multiple amplified products and can be implemented on a portable device for accurate infectious disease screening in resource-limited settings.

Screening for gene doping transgenes in horses via the use of massively parallel sequencing

Jillian Maniego et al.

Summary: Genetic manipulation is predicted to be the next frontier in doping, with massively-parallel sequencing allowing for screening of multiple doping targets simultaneously. The study demonstrated the high scalability and sensitivity of parallel sequencing by comparing its detection sensitivity with real-time qPCR in a real-world setting involving intramuscular administration of a control rAAV vector into two horses. The results showed that both methods detected the vector up to 79 hours post-administration, with sporadic detection after 96 hours.

Detection of non-targeted transgenes by whole-genome resequencing for gene-doping control

Teruaki Tozaki et al.

Summary: This study aimed to detect genetically engineered animals for gene-doping control using whole-genome resequencing techniques, successfully identifying transgenes as intron deletions. By conducting genome-wide screening, artificially introduced single-nucleotide polymorphisms and insertions/deletions in transgenes were identified.

Robustness of Digital PCR and Real-Time PCR in Transgene Detection for Gene-Doping Control

Teruaki Tozaki et al.

Summary: The study investigated the robustness of digital PCR and real-time PCR in transgene detection. Digital PCR was relatively robust when primer and probe sequences did not completely match the template transgene, while real-time PCR was less sensitive. Lowering the annealing temperature can improve PCR sensitivity.

Genome-wide programmable transcriptional memory by CRISPR-based epigenome editing

James K. Nunez et al.

Summary: CRISPRoff is a programmable epigenetic memory writer that can heritably alter gene expression by initiating specific DNA methylation and gene repression. The tool utilizes a single dead Cas9 fusion protein to establish these modifications, which are maintained through cell division and differentiation processes. By pairing CRISPRoff with genome-wide screens and analysis of chromatin marks, rules for heritable gene silencing can be established, with the tool capable of targeting a wide range of genes beyond canonical CpG islands.
