Temperature Regulates Stability, Ligand Binding (Mg2+ and ATP), and Stoichiometry of GroEL-GroES Complexes

Thomas E. Walker et al.

Summary: The study used variable-temperature electrospray ionization native mass spectrometry to analyze the stability of GroEL and GroEL-GroES complexes, revealing destabilization at lower temperatures and the formation of previously unreported reaction products. Additionally, it was found that temperature, Mg2+, and ATP concentrations influence the stoichiometry of the GroEL-GroES complex.

Andreas Walz et al.

Summary: The prototype of a highly versatile and efficient preparative mass spectrometry system used for the deposition of molecules in ultrahigh vacuum (UHV) is presented. The system combines three designs of innovative ion guides, a novel digital mass-selective quadrupole (dQMF), and a standard electrospray ionization (ESI) source to achieve full control. The system demonstrates encouraging performance in terms of resolution and overall efficiency.

Simulations of a digital ion filter and a digital ion trap for heavy biomolecules

Florian Simke et al.

Summary: Ion trajectories were simulated for a linear quadrupole ion-filter and a linear Paul trap in order to study the biomolecules using single-particle imaging with X-ray pulses. The analysis focused on the transmission efficiency, mass resolving power of the filter, and ion capture and thermalization in the trap. The resulting optimized ion bunch delivered by the assembly was characterized.

THE IMS PARADOX: A PERSPECTIVE ON STRUCTURAL ION MOBILITY-MASS SPECTROMETRY

Jacob W. McCabe et al.

Summary: Studies on large proteins, protein complexes, and membrane protein complexes present new challenges in terms of increased ion mobility and mass spectrometry resolution. The development of IM-MS techniques has shown promising results in analyzing complex biomolecular structures and retaining solution phase conformations in gas phase ions. The integration of Fourier-transform IM-MS instruments has revolutionized the field by providing first-principles determinations of collision cross sections and enhancing the capabilities of native MS/IM-MS techniques.

Quantifying the operation of sinusoidal mass filters

Adam P. Huntley et al.

Summary: This manuscript systematically quantifies the effects of DC and AC voltages on resolution, resolving power, pseudopotential well depth, and transmission in sinusoidal quadrupole mass filters. Commercial quadrupole instruments rarely scan at constant resolving power, instead opting for constant resolution, where the mass window width is fixed. Commercial mass filter systems achieve constant resolution scans by linearly changing DC and AC voltages at a fixed ratio, with an additional negative DC voltage offset.

Entropy in the Molecular Recognition of Membrane Protein-Lipid Interactions

Pei Qiao et al.

Summary: This study used variable-temperature native mass spectrometry to investigate the thermodynamics of lipid binding events to the human G-protein-gated inward rectifier potassium channel, Kir3.2, revealing distinct thermodynamic strategies for different types of phosphatidylinositol and phosphorylated forms. The acyl chain of the lipid and the number of phosphate groups have significant impacts on the binding thermodynamics, with entropy and enthalpy playing key roles in the interactions.

Summary: This study presents a digital-waveform dual-quadrupole mass spectrometer to improve the performance of a drift tube FT-IMS high-resolution Orbitrap mass spectrometer in studying large proteins and protein complexes. The dual-quadrupole analyzer allows for low-energy collisional activation of ions and precise mass selection using digital-waveform technology, enhancing the capabilities of the instrument at a lower cost compared to traditional RF drivers.
