Single-cell sorting using integrated pneumatic valve droplet microfluidic chip

Summary: Single-cell analysis and sorting is an important technique in scientific research and industry. This study developed a microfluidic chip that integrates single-cell droplet generation, incubation, and sorting functions using a pneumatic micro-valve. Summary: In this study, a digital loop-mediated isothermal amplification (LAMP) assay was developed for counting hepatitis B virus (HBV) DNA using single-particle (sp) inductively coupled plasma mass spectrometry (ICP-MS). By intercalating LAMP amplicons into agarose particles and labeling them with [Ru(bpy)(2)dppz](2+), the agarose particles containing virus DNA could be detected in spICP-MS. This method enables quantitative analysis of extremely low concentrations of virus DNA without the need for a calibration curve.
