On-line Sequencing of CRISPR Guide RNAs and Their Impurities via the Use of Immobilized Ribonuclease Cartridges Attached to a 2D/3D-LC-MS System

Summary: This study introduced for the first time the automated digestion and sequencing of an RNA molecule using immobilized RNase cartridges attached to a multidimensional LC-MS system. The on-line approach showed higher efficiency and lower sample requirement compared to the conventional off-line approach. The study also developed a three-dimensional LC-MS method for sequencing fractions collected on-line in order to gain insights into the cause of main peak tailing.

Summary: With the development of RNA-based therapeutics, the need for analytical methods to confirm sequences and map modifications has become urgent. This study compares different mass spectrometry techniques for sequencing modified nucleic acids and finds that UV photodissociation and activated electron photodetachment dissociation are effective for characterizing modified nucleic acids. However, different modifications can affect the fragmentation pathways and sequence coverage.
