Embossed Paper Platform for Whole Blood Collection, Room Temperature Storage, and Direct Analysis by Pinhole Paper Spray Mass Spectrometry

Summary: Dry-state microsampling techniques, based on embossed hydrophobic paper substrate, provide a convenient and advantageous method for sample collection in resource-limited settings. In this study, a new pinhole paper spray ionization method was developed for direct mass spectrometry analysis of dried blood samples, offering a rapid and efficient alternative to the traditional liquid chromatographic-mass spectrometry approach. The sensitivity and sample stability were found to be superior in the embossed hydrophobic paper compared to the planar hydrophilic paper.

The hLMJ-SSP was controlled by a microcontroller unit and allowed for automated surface analysis within about 2 minutes. The system achieved a low limit of detection and demonstrated its capability for in-situ analysis of illicit drug residues and medicines in biological fluids.
