Extraction of intrinsic column peak profiles of narrow-bore and microbore columns by peak deconvolution methods

Summary: In this study, intrinsic peak profiles generated by narrow-bore and microbore chromatographic columns in LC-MS proteomic analyses were extracted using two different deconvolution methods. The first method, based on classical discrete Fourier transform (DFT), is most relevant when the low-frequency band of the chromatographic peak does not overlap with high-frequency noise bands. The second method, relying on Taylor expansion of continuous Fourier transform (FT), is successful for extracting intrinsic peak profiles of narrow-bore columns packed with sub-2 mm particles on standard UHPLC systems.
