UHPLC-ESI-MS/MS assay for quantification of endocannabinoids in cerebrospinal fluid using surrogate calibrant and surrogate matrix approaches

Summary: Endocannabinoids are endogenous lipids that act as neuromodulators through cannabinoid receptors. This study developed a sensitive UHPLC-MS/MS method for the analysis of trace levels of 7 endocannabinoids in cerebrospinal fluid samples. The method demonstrated excellent sensitivity, accuracy, and reliability, and was used to quantify endocannabinoids in 118 human cerebrospinal fluid samples. Differences in endocannabinoid concentrations were observed between patients with CNS infection and controls.

It has been demonstrated to evaluate differences in steroid content between peripheral serum and peritoneal fluid across menstrual cycle phases and the effect of the synthetic gestagen dienogest on the steroid metabolome.

Internal calibration as an emerging approach for endogenous analyte quantification: Application to steroids

Summary: In this study, an innovative internal calibration approach using stable isotope-labeled standard version of endogenous compound was developed to improve the quantitative performance of mass spectrometry methods. The results showed that the response factors obtained on different LC-MS platforms were consistent and the quantitative results in human serum samples were similar to those obtained with conventional quantification approach based on external calibration. Furthermore, it was demonstrated that the naturally occurring isotopes of SIL could be used to increase the accuracy for analytes with low concentrations.

A total of 161 metabolites were successfully detected in ESI+ mode, and 92 were detected in negative ionization mode. The developed methods were calibrated and validated, showing good accuracy and precision.

The study developed a new microflow UHPLC-ESI-MS/MS method for measuring salivary cortisol and cortisone, showing enhanced sensitivity and eco-friendliness. Surrogate calibrant approach with labeled internal standards was used for quantification, and the method was successfully validated for routine clinical stress studies with over 4000 injections.
